摘 要:背景与目的:低氧诱导因子是广泛存在于多种细胞的转录因子,在低氧时肿瘤细胞相关基因的表达发挥着重要的调控作用。本研究探讨了低氧诱导因子HIF-1α mRNA和HIF-2α mRNA在肾透明细胞癌中的转录及其临床意义。方法:采用逆转录多聚酶链式反应(RT-PCR)方法检测56例肾透明细胞癌组织、对照组43例远离肿瘤的癌周肾脏组织及9例良性疾病肾脏组织中HIF-1α mRNA和HIF-2α mRNA的阳性率。结果:HIF-1α mRNA在肾透明细胞癌组织中的阳性率为87.5%,较对照组织13.5%高,差异具有显著性(P<0.001);HIF-2α mRNA在肾透明细胞癌标本组织中的表达率为94.6%,较对照组织7.7%高,差异有显著性(P<0.001)。结论:HIF-1α mRNA和HIF-2α mRNA的转录可能对肾透明细胞癌的发生和发展有重要作用。Background and purpose: Clear cell renal cell carcinoma is the most common type of malignant renal tumor. Hypoxia inducible factor has been reported to be expressed in many types of cells, and plays a key role in modulation of tumor related genes under hypoxia. Our research was to investigate the clinical value of hypoxia inducible factor-1α (HIF-1α) and hypoxia inducible factor-2α(HIF-2α) mRNA expression in clear cell renal cell carcinoma (CCRCC). Methods: RT-PCR was used to examine the expression level of HIF-1αand HIF-2αmRNAs in the tumor tissues from 56 patients with CCRCC. The control group included the tissues of benign real diseases and the normal tissues distant from the tumor in the kidney. Results: Compared with the control group, the expression level of HIF-1αand HIF-2αmRNA in clear cell renal carcinoma was much higher( HIF-1α: 87.5% vs. 13.5% ; HIF-2α: 94.6% vs. 7.7% ) and there was a significant difference between tumors and the control group( P =0. 001 6). Conclusions: HIF-1αand HIF-2α may be important factors for the development and progression of clear cell renal cell carcinoma.
