摘  要:目的研究免疫抑制剂对白细胞介素-2(IL-2)诱导大鼠急性癫痫的抑制作用,及其对大脑皮质及海马内谷氨酸(Glu)I、L-2受体(IL-2R)表达的影响,探讨免疫抑制剂抗痫效应及其作用机制。方法将大鼠随机分为4组:A组(生理盐水对照组)、B组(IL-2组)、C组(环孢素-A+IL-2组)、D组(糖皮质激素处理组),对大鼠行侧脑室注射相应试剂后观察并记录大鼠行为表现,采用免疫组织化学方法检测大脑皮质及海马内Glu的含量及IL-2R的表达。结果A组无明显痫样发作;B组痫样发作程度达Ⅲ级;C组发作程度为Ⅰ级;D组无痫样发作。免疫组化染色显示,各组注药后2h,B组的大脑皮质及海马内GluI、L-2R免疫反应较A组明显增强,差异具有显著性意义。C组与A组相比,GluI、L-2R免疫反应无明显变化。D组与A组相比,GluI、L-2R免疫反应减弱。结论IL-2具有致痫效应,免疫抑制剂具有一定的抗痫作用,其机制与抑制Glu和IL-2R表达有密切关系。Objective To explore the anti-epileptic effect of immunosuppressants of glutamate (Glu) and interleukin-2 receptor (IL-2R) of cerebral cortex and hippocampal. Methods The rats were randomly divided into 4 groups: group A, normal saline control group; group B, IL-2 group; group C, cyclosporine (CsA)+IL-2 group; group D, glucocorticoid (GC) group. The behavior was observed after injecting IL-2 into the lateral ventricle of rats and the expression of Glu and IL-2R of cerebral cortex and hippocampal was detected by using immunohistochemical staining. Results There was no epilepsy in groups A and D. The rats in group B showed epileptic activities about gradeⅢ. The seizure was partly restrained in rats of group C, showing grade Ⅰ epileptic activities. The immunohistochemical staining revealed that, 2 h after injection, the immunoreactive (IR) positive cells of Glu and IL-2R in cerebral cortex and hippocampal of group B were significantly increased as compared with group A. There was no significant difference in IR positive cells of Glu and IL-2R between group C and group A. Conclusion IL-2 could induce the epileptic seizure. The immunosuppressants could restrain the seizure, and its mechanism might be related to the inhibition of the expression of Glu and IL-2R.
