摘 要:目的研究全反式维甲酸对人骨肉瘤细胞凋亡的影响。方法在培养液体系中加入不同浓度的全反式维甲酸(ATRA)进行不同时间长度的诱导,以MTT法,流式细胞术,TUNEL法检测细胞凋亡情况。结果1~50μmol/LATRA均能抑制骨肉瘤细胞增殖。5、10、50μmol/LATRA作用细胞4d后的凋亡率分别为(12.46±2.37)%、(18.36±1.32)%、(27.15±2.17)%,与对照组[(4.09±1.18)%]比较差异均有显著性意义(均P<0.05)。ATRA浓度越高,抑制作用越明显,呈明显的剂量依赖效应。50μmol/LATRA作用细胞1、2、3、4d后的凋亡率分别为(7.35±1.28)%、(10.72±1.68)%、(17.65±2.14)%、(27.15±2.17)%,与对照组[(4.09±1.18)%]比较差异均有显著性意义(均P〈0.05)。ATRA作用时间越长,抑制作用也越明显,呈明显的时间依赖效应。结论ATRA能够诱导人骨肉瘤细胞发生凋亡,呈时间和剂量依赖效应。Objective To investigate the effects of all-trans retinoic acid (ATRA)-induced apoptosis of human osteosarcoma cells. Methods Different concentrations of ATRA were added into culture media system to induce apoptosis of human osteosarcoma cells. The apoptosis was detected by MTT assay, flow cytometry and TUNEL method. Results ATRA at concentrations of 1- 50 μmol/L could inhibit the proliferation of osteosarcoma cells in a time- and dose-dependent manner. After the cells were treated with 5, 10 and 50 μmol/L ATRA for 4 days, the rates of apoptosis were (12.46±2.37) %, (18. 36± 1.32) % and (27.15±2. 17) % respectively, showing significant difference with that in control group [(4. 09 ± 1.18)%, P〈0. 05]. The rates of apoptosis in the osteosarcoma cells induced by 50 μmol/L ATRA for 1, 2, 3 and 4 days were (7.35 ± 1.28)%, (10. 72 ± 1.68) %, ( 17.65± 2.14) % and (27.15±2.17) % respectively, significantly higher than in control group E(4.09± 1.18) %, P〈0. 05], showing a time-dependent manner. Conclusion ATRA could induce apoptosis in human osteosarcoma cells in a time- and dose-dependent manner.
