摘  要:目的:观察人多药耐药基因mdr1启动子调控的胞嘧啶脱氨酶-尿嘧啶磷酸核糖转移酶融合自杀基因(CD::upp)对紫杉醇耐药卵巢癌裸鼠皮下移植瘤的抑制作用及对荷瘤裸鼠生存时间的影响。方法:建立卵巢癌耐药及非耐药细胞的裸鼠模型,将含mdr1-CD::upp的重组腺病毒0.1ml(1×10^9pfu/ml)注入裸鼠皮下移植瘤体内,腹腔内注射5-氟胞嘧啶(5-FC),观察各组裸鼠肿瘤生长速度及存活时间。结果:实验组裸鼠移植瘤明显受到抑制,与对照组的差异有统计学意义(P〈0.05)。实验组荷瘤裸鼠的生存时间明显延长,对照组在69天内全部死亡,差异有统计学意义(P〈0.001)。结论:mdr1启动子调控的CD::upp基因能特异性地抑制卵巢癌裸鼠耐药移植瘤的生长并延长生存时间。Objective:To evaluate the anticancer effects of adenovirus-mediated CD :: upp gene directed by mdr1 promoter on drug-resistance ovarian cancer in vivo. Methods:Adenovirus liquid containing mdr1-CD :: upp gene was directly injected into tumor xenografts of nude mice and 5-FC administration was followed. The overall survival and anti-tumor effect was observed. Results: Compared with the controlled group, the growth of tumors in tested group was signifiicanfly suppressed, and there was a significant difference ( P 〈 0.05 ). The survival of the tested group was prolonged compared with the controlled group ( P 〈 0. 001 ). Conclusion: Transduction of this adenovirus vector containing mdr1-CD :: upp resulted in suppression of tumor growth and prolonged survival in drug-resistance tumor-bearing nude mice.
