胰腺癌组织中CDC25B表达及临床意义的初步探讨

摘　　要：目的研究CDC25B基因在胰腺癌组织中的表达,探讨其在胰腺癌发病机制中的作用。方法利用实时荧光定量-PCR技术检测24例胰腺癌组织和4例正常胰腺组织中CDC25B基因的表达,PCR产物的定量方法采用比较Ct法,应用western blot检测其蛋白的表达水平。用Kaplan-Meier生存率曲线分析CDC25BmRNA与预后的关系。结果实时荧光定量-PCR发现CDC25B基因平均模板量在正常胰腺组织中为0.000635±0.000210,胰腺癌组织中为0.001988±0.000650;与正常胰腺组织相比,胰腺癌组织中CDC25B mRNA表达上调3.13倍(P<0.05),CDC25B mRNA升高5倍以上者生存时间缩短。胰腺癌组织中CDC25B蛋白也过度表达,正常胰腺组织CDC25B蛋白表达为1.22±0.33,胰腺癌组织为5.56±1.57,上调4.56倍(P<0.01)。结论CDC25B在胰腺癌组织中表达明显上调,CDC25B mRNA升高者预后差。Objective： To investigate the expression of cell division cycle 25 B gene （CDC25 B） in pancreatic adenocarcinoma tissues and its role in the pathogenesis of pancreatic cancer. Methods：Tbe expression of CDC25 B gene in 24 cases with pancreatic cancer and 4 cases of normal pancreas were evaluated by real-time fluorogenetic quantitative PCR （RFQ-PCR）. The product of RFQ- PCR was quantitated by comparative Ct method. The protein level of CDC25B was determined by Western blot. The association between expression of CDC25B mRNA and prognosis of patients were analyzed by Kaplan-Meier survival curve. Results： QRT-PCR showed that the average quantity of CDC25B mRNA in pancreatic adenocarcinoma and normal pancreas was 0.001 988 ±0.000 65 and 0. 000 6350 ± 0.000 21, respectively. In comparison with normal pancreas, the expression of CDC25B gene in pancreatic adenocarcinoma had 3.13 fold up-regnlation （P 〈 0.05 ）. The patients with 5-fold increase in CDC25B gene expression had shorter survival time. Western blot revealed that compared with normal pancreas CDC25B protein had 4.56 fold increase in pancreatic adenocarcinoma tissues （ 1.22 ± 0.33 vs 5.56 ± 1.57 ）. Conclusion： CDC25B was overexpressed in pancreatic adenocarcinoma tissues. Patients with overexpression of CDC25B mRNA had poor prognosis.
