摘  要:目的:研究阿司匹林在健康人药效学特征。方法:选择健康汉族男性志愿者28名,随机分成阿司匹林3个剂量组: 81mg组(n=9)、162mg组(n=9)及324mg组(n=10),每日1次,连续服用7天,在服药前0h、服药后2h、24h、96h及停药后1、3、7天,分别测定血中血栓素B_2(TXB_2)和血小板聚集率(PAg)。结果:与服药前比较,在服药后2h,三组TXB_2、PAg抑制率基本达到了最大,TXB_2生成持续受抑制维持至停药后3天,而三组PAg抑制率在服药24h、96h均有所升高,在服药7天后,三组PAg抑制率恢复至服药后2h水平。结论:单次口服阿司匹林81~324mg,抑制血小板聚集作用在2h基本达到最大,且无剂量间差异;连续服药时,三组阿司匹林抑制TXB_2作用相同,连续服药7天后,抑制血小板聚集作用达到稳定,停药3天后血小板功能恢复正常。Objective: To evaluate aspirin pharmacodynamics in healthy volunteers. Methods: 28 volunteers were chosen from Chinese healthy male subjects and randomly assigned to three test subgroups: 81mg(n=9), 162mg(n=9) and 324mg(n=10). The test drug was aspirin, 8 lmg per tablet. All volunteers were orally administered everyday for successive 7 days. Thromboxane B2 (TXB2) and Platelet aggregation (PAg) were analyzed before and after oral administration. The samples of TXB2 and PAg were collected before medication in the day 1(0h) and after oral application at 2h, 24h, 96h and day 1, 3, 7 after drug withdrawal. Results: TXB2 and PAg were maximally inhibited at 2 hours after oral administration in three subgroups. TXB2 was inhibited continuously till 3 days of drug withdrawal. But PAg was increased after 24h and 96h in three groups, it was maximally inhibited after successive 7 days again. Conclusions: After 2 hours of single oral application, TXB2 and PAg had been maximally inhibited by aspirin, there was no difference among three groups. There were same effects on TXB2 among three groups in successive medication. The inhibitive effect on PAg became stable after successive 7 days. After 3 days of drugs withdrawal, TXB2 and PAg returned to normal level.
