摘 要:目的以时间治疗学的原理为基础对放射治疗进行改良,以获得更佳的疗效。方法首先将肿瘤细胞LLC细胞悬液接种至已接受2周光暗适应的雄性C57BL/6J小鼠皮下。待肿瘤长至直径1.0厘米时,按特定时间点分组(组照射时间11HALO,组照射时间为23HALO)对小鼠行^(60)Co-γ射线一次性全身照射。之后观察各组肿瘤大小,并对肿瘤组织行HE染色和PCNA免疫组化染色,进而观察肿瘤细胞形态和增值情况。结果组较组而言,肿瘤再生长的速度更加缓慢,且出现更明显的细胞退行性变和区域性坏死,同时PCNA表达量明显降低。结论在11HALO行放射治疗的小鼠肿瘤生长明显比23HALO行放射治疗的小鼠和未经放射治疗的小鼠缓慢,肿瘤组织受损也相对严重。说明在该时间点行放射治疗有助于提高放疗的效果。Objective To improve the radiotherapeutic method with chronotherapeutics principle and thereupon contribute favorably to the therapeutic effect. Methods LLCs were cultured, harvested, made into ceil suspension and planted into male C57BL/6J mice which had been raised in 12h light and dark cycles for 2 weeks. The mice were divided into different groups and exposed to ^60Co-γ-ray at certain time (Group Ⅰ at 11HALO, Group Ⅱ at 23HALO, HALO: hour after light on). Then the tumor size was observed, HE Stain and PCNA immunohistochemistry were performed to detect the tumor ceils appearance and generation of different groups. Results Comparing with Group Ⅱ and control group, the tumor regrowth-speed was slower in Group Ⅰ , and tumor cell degeneration, regional cellular necrosis were more conspicuous and PCNA expression was lower in Group Ⅰ . Conclusion Tumors exposed to llHALO can retard their growth. Radiotherapy at llHALO is helpful for improving curative effect.
