摘  要:目的研究葡萄籽提取物(GSE)对四氯化碳(CCl4)诱导肝损伤的抗氧化保护作用。方法建立CCl4诱导肝损伤的大鼠模型,测定正常对照组、CCl4诱导组(CCl4组)、GSE低剂量组(20mg/kg·d)、GSE中剂量组(40mg/kg·d)和GSE高剂量组(120mg/kg·d)共5个组肝脏中总抗氧化能力(TAOC)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)水平和血清SOD、TAOC水平,称量肝重和体重,比较各组大鼠肝体比值、甘油三酯(TG)的改变。结果各组大鼠肝脏中TAOC值和SOD值差异无统计学意义;CCl4组和GSE低、中剂量组的肝脏中GSH-Px值高于对照组和GSE高剂量组;CCl4组肝脏MDA值组显著高于正常对照组及GSE低、中、高3个剂量组。GSE高剂量组的血清TAOC值显著高于其余4个组;高剂量组、中剂量组血清中SOD显著高于其余3组。5个组的肝体比值无统计学意义;CCl4组的肝脏中TG值高于正常对照组和GSE中剂量组。结论葡萄籽提取物可降低大鼠肝脏中TG和MDA含量,提高机体总抗氧化力,明显改善肝脏病理改变。Objective To study the anti-oxldation effects of grape seed extract (GSE) on carbon tetrachloride (CCl4) induced hepatic injury. Method Established CCl4 induced hepatic injury on rats. Hepatic total anti-oxidation capacity (TAOC), superoxide dismutase (SOD), glutathion peroxidase ( GSH-Px ), malondialdehyde (MDA) concentrations and blood SOD, TAOC concentrations were measured in CCl4 group, low dose (20 mg/kg·d) GSE group (LDG group), medium dose (40 mg/kg·d ) GSE group (MDG group), high dose (120 mg/kg·d) GSE group (HDG group), and normal group (was not given CCl4 and GSE). We also determined the hepatic weight to body weight ratio. TG level measured were checked through microscope. Result There were no significant differences between five groups on hepatic TAOC and SOD value ; compared with normal group and HDG group. Hepatic GSH-Px values in CCl4 group, LDG group and MDG group were higher. Hepatic MDA value in CCl4 group was higher than the other four groups. HDG group's blood TAOC value was higher than the other four groups; blood SOD values in MDG group and HDG group were higher than the other three group. No significant differences were observed on the hepatic weight to body weight ratios between five groups. CCl4 group's hepatic TG content was significantly higher than normal group and MDG group. Conclusion Grape seed extract can lower contents of hepatic TG and MDA, increase total antl-oxidation capacity and obviously improve hepatic pathological change.
