摘 要:目的观察N-乙酰半胱氨酸滴眼液是否能延缓链脲佐菌素(streptozotocin,STZ)诱导的大鼠糖尿病性白内障。方法选择85只sD大鼠,随机抽取lO只为正常组,其余75只大鼠采用一次性腹腔内注射STZ(65mg·kg^-1)的方法建立大鼠糖尿病模型,血糖值大于14mmol·L^-1被判定为糖尿病大鼠,并完全随机化分成3个组(B组18只,C组19只,D组19只)。A组为正常对照组,未给予任何处理;B组和c组为糖尿病治疗组,分别给予O.1g·L^-1与0.5g·L^-1的N-乙酰半胱氨酸溶液滴眼,均为每日2—3次;D组为糖尿病对照组,给予磷酸钠缓冲液滴眼。每周监测大鼠尿糖、体重的变化,并在裂隙灯下观察大鼠晶状体混浊的进展;每月监测大鼠的血糖变化。结果78%(59/75)大鼠达到成模标准,随机血糖大于14mmol·L^-1,并出现糖尿病表现。糖尿病大鼠晶状体混浊呈2阶段发展,前6周缓慢进展及后7周快速进展。N-乙酰半胱氨酸滴眼液在治疗的前5周可延缓糖尿病性白内障的的发生发展,其中第4周出现统计学差异(P〈0.05);而在治疗的后期,各糖尿病组的晶状体混浊程度明显加重,并且各组之间进展程度没有明显差异。结论N-乙酰半胱氨酸滴眼液能安全有效的延缓早期糖尿病性白内障的发生发展。[眼科新进展2007;27(3):176—178]Objective To observe whether N-acetylcysteine drops can delay the dtaheUc cataract induced by streptozotoc (STZ) in rats. Methods Eightyflve SD rats were selected, 10 rats were set randomly as normal group A, and others were injected STZ( 65 mg· kg^-1) to establish the diabetic model, the rats with blood glucose more thnn 14 mmol· L^-1 were assessed as diabetic rats.Then 56 dtabetic rats were divided into three groups,18 rats in group B,19 rats in group C,19 rats in group D;Group B and C set as diabetic treatment . group and group D as dtabetic control group. The rats in group A receded no treated,group B and C received 0. 1 g· L^-1 and 0.5 g· L^-1 N-acetylcysteine drops respectively, and group D received sodium phosphate bnlaneed soluUon. The change of urine glucose and body weight were monitored and transparence of lens was obseved by slit lamp biomicroscope weekly. The change of blood glucose was also monitored monthly. Results The random blood gluocose in 59 rats was more than 14 mmol · L^-1 and the diabetic symptom appeared in these rats. Lens opactification progressed in a biphasic manner in diabetic rats,an inisial slow increase in the first 6 weeks, then a steep increase in the next 7 weeks. N-aeetylcystetne drops delayed the progression of dtabetic cataract in the first 5 weeks,and there was significant difference on the 4th week( P 〈0.05). In the last stage of treatment, the degree of lens opaeification in each diabetic group was aggravated, and there was no difference among these groups. Conclusion N-acetylcysteine drops can delay the progression of diabetic cataract at the ear ly stage safely and effectively. [ Rec Adv Ophthaimol 2007 ;27 (3) : 176-178 ]
