摘 要:目的探讨宫颈浸润癌患者与健康女性间血清中的蛋白质差异,以寻找评价治疗效果和判断预后的生物学指标,为制定合理的治疗方案寻找依据。方法通过表面增强激光解吸离子化飞行时间质谱仪(sur-face enhanced lazar desorption/ionisation-time of flight-mass spectrometry,SELDI-TOF-MS),铜离子结合芯片(immobilized metal affinity capture arrays-Cu,IMAC-Cu)以及配套软件,检测49例宫颈浸润癌,71例年龄相配的健康女性血清,筛选出有分类意义的差异蛋白,寻找出合理的运算规则,构建决策树分类模型。结果宫颈浸润癌与正常对照组比较,共有47种蛋白质质谱峰差异有显著性(P<0.05),有分类意义的蛋白质有6种,质荷比为M8929.31,M7930.52,M9127.31,M8141.01,M7963.06,M9280.63,在宫颈浸润癌患者血清中低表达,含量明显低于正常对照,用质荷比为M8929.31的蛋白质建立决策树分类模型,分类正确率为98.33%(118/120),敏感性为97.96%(48/49),特异性为98.59%(70/71)。结论质荷比为M8929.31,M7930.52,M9127.31,M8141.01,M7963.06,M9280.63的一组蛋白质与宫颈浸润癌密切相关,有可能成为评价宫颈癌治疗效果以及预后判断的一组生物学指标。Objective To compare the significant serum proteins in patients with cervical invasive cancer and age - matched healthy women. To seeking for a group of serum biomarkers that can be used to monitor the treatment and evaluate the prognosis of patients with cervical cancer. Methods Surface enhanced lazar desorption/ionisationtime of flight - mass spectrometry ( SELDI - TOF - MS ) with immobilized metal affinity capture arrays - Cu ( IMAC - Cu) was used to examine the serum of 49 patients with cervical invasive cancer and 71 healthy women. The data of proteomic spectra were analysed by a Biomarker pattern software and Biomarker Wizard software to build a diagnostic model by a decision tree classification algorithm with 10 - fold cross validation. Results A total of 47 types of protein presented in significantly different intensity (P〈0. 05) between the sera of cervical invasive cancer patients and control. Six proteins with the ratio of mass to charge (M/Z) of M8929.31, M7930.52, M9127. 31, M8141.01, M7963.06 and M9280. 63 were of importance as decision tree classification algorithm, which appeared to be down regulated in sera of patients with cervical invasive cancer. The decision tree model of the protein with M/ Z of M8929.31 is with 98.33% (118/120) of correct classification ratio, 97.92% (47/48) sensitivity, 98. 59% (70/71) specificity. Conclusions The group of biomarkers with M/Z of M8929.31, M7930. 52, M9127.31, M8141.01, M7963.06 and M9280. 63 might be potentially valuable in monitoring of therapy response, and predicting the prognosis of cervical cancer.
