摘  要:为了比较两种方法测定昆虫抗冻蛋白生物学活性的优劣.通过抗冻蛋白细菌低温保护实验测定菌液OD500和菌落计数两种方法同时研究洛氏脊漠甲抗冻蛋白对大肠杆菌的低温保护效果,实验表明发现这两种方法所得结果相近,相关系数r=0.82~1.0,相关性显著(p〈0.05).菌落计数法的误差小NOD值法,适用于菌落数少的情况,OD值法适用于菌落密度大的情况,两种方法都可用于研究抗冻蛋白低温保护活性.To compare two methods for measuring the biological activity of antifreeze protein, cryoprotective experiment was employed to check the protective effect of insect antifreeze protein from Pterocoma loczyi on bacteria at subzero temperature by measuring the value of OD500(OD-measuring) and counting the number of the bacterial colonies (colony-counting) separately after the antifreeze protein was added to bacterial culture and stored at -7℃ for various hours. The results showed that the two methods produced similar cryoprotective effect on bacteria. The correlation coefficient ranges from 0. 82 to 1.0 (p〈0. 05). The experiment error in colonycounting is less than that in OD-measuring. Both methods can be used to study the cryoprotective activity of antifreeze protein, but colony-counting is more accurate and suitable for lower colony density, while OD-measuring more convenient for higher colony density of bacteria of the culture. Both methods can be applied to study the biological activity of antifreeze protein for protecting bacteria at subzero temperature.
