摘  要:目的研究谷氨酰胺(Gln)对体外培养肝癌细胞株SMMC-7721内热休克蛋白70(HSP70)表达的影响,及其与细胞增殖的关系。方法SMMC-7721细胞株分别与0、0.5、1.0、2.0、4.0、8.0和16.0mmol/L的Gln培养24小时后,采用MTT法检测SMMC-7721细胞的增殖,RT-PCR和Western blot方法检测细胞内HSP70 mRNA和蛋白质的表达。结果Gln能显著促进SMMC-7721细胞增殖,当Gln浓度为2.0mmol/L时,SMMC-7721细胞的增殖达峰值。SMMC-7721细胞内有HSP70表达,Gln显著增加HSP70 mRNA和蛋白质的表达(P<0.05),当Gln浓度为2mmol/L时,HSP70表达量最高。结论诱导SMMC-7721细胞HSP70表达增高可能是Gln促肿瘤生长的作用机制之一。Objective To evaluate the influence of glutamine (Gin) on heat shock protein 70 (HSP70) expression in hepatoma cell line SMMC-7721 in vitro, and to investigate its relationship with cell proliferation. Methods After SMMC-7721 cells were cultured with 0, 0. 5, 1.0, 2.0, 4.0, 8.0, and 16.0 mmol/L Gin for 24 hours respectively, cell proliferation was evaluated by MTT method, and HSP70 mRNA and protein expressions were examined using RT-PCR and Western blot, respectively. Results Gin promoted SMMC-7721 cell proliferation significantly. The expressions of HSP70 mRNA and protein in SMMC-7721 cells were significantly elevated by Gin treatment ( P 〈 0. 05 ). Both the proliferation and the HSP70 expression reached the peak at a Gin concentration of 2.0 mml/L. Conclusion The induction of HSP70 expression possibly is one of the mechanisms of Gin contributing to hepatoma cell growth.
