摘 要:研究使用双歧杆菌免疫家兔和小鼠,制备高效价抗兔、抗鼠免疫血清的方法。将双歧杆菌厌氧培养后,制成1010mL-1活的菌悬液,皮下注射免疫家兔和小鼠,采用试管凝集法检测血清效价,采用ELISA法确定双歧杆菌的最低检测浓度。结果表明,经试管凝集法检测血清效价,效价达到1:5120;制备的双歧杆菌免疫血清与双歧杆菌属的菌株有很强的交叉反应,与发酵乳制品中常见的细菌未见明显交叉反应;经改良双夹心ELISA法检测,对双歧杆菌的最低检测浓度为105mL-1。制备了高效价的双歧杆菌免疫血清,这一结果为双歧杆菌乳制品中双歧杆菌的血清学检测提供了参考。The methods to produce high-titer anti-Bifidobacterium serum by immunizing rabbits and mice with Bifidobacterium were investigat- ed. All rabbits and mice were vaccinated with 1×10^10 cfu/mL live Bifidobacterium suspension which were made from B. longum ATCC 15707, B. infant KLDS 2.0505 and B. bifidum KLDS 2.0503 culture by subcutaneous injection. Serum efficiency was detected by tube agglutination test, and the detection limit for Bifidobacterium by the immune serum was determined by Enzyme-linked Immunosorbent Assay (ELISA). (1) The agglutination efficiency of the immune serum reached 1 : 5120; (2) The immune serum was Bifiodobacterium species specific, and reaction with non-Bifidobacterium species was not observed; (3) The detection linfit for Bifidobacterium by the immune serum was 10^5 cfu/mL. Hightiter anti-Bifidobacterium serum with Bifidobacterium species were produced in this study, and these results provide a strategy to detect Bifidobacterium in dairy products serologically.
