不同中医治法对实验性脑出血大鼠Caspase-3表达的影响

摘　　要：目的观察大鼠脑出血后血肿周围神经元活化Caspase-3的表达情况及不同中医治法的干预作用。方法用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型,免疫组化检测血肿周围活化Caspase-3的表达。结果大鼠脑出血后血肿周围神经元于6h出现Caspase-3阳性表达细胞,1d阳性细胞计数达高峰,3d出现轻度下降,5d明显下降,与假手术组比较差异明显(P<0.01),采取不同中医治法治疗后有所下降,但仍高于假手术组;与其他各组比较,平肝熄风汤组治疗后下降最明显(P<0.01)。结论脑出血后血肿周围神经元活化Caspase-3表达上调,采取熄风、泻火、祛痰、化瘀以及风火痰瘀同治等治法均能抑制Caspase-3的活化,阻止神经元的凋亡,以平肝熄风汤组最为明显。Objective To observe the effects of different traditional Chinese medicine on the expression of activated Caspase-3 in neuron around hematoma and the changes of the hematoma after intracerebral hemorrhage（ICH） in rats. Methods Type Ⅶ collagenase was stereotaxically injected into the globas pallidus of rats to establish the experimental ICH models. Immunohistochemical staining was used to examine the expression of activated Caspase- 3 after ICH in rat at different time points. Results The positive cells were appeared around the hematoma at 6h after operation and peaked to the top at ld, then declined slightly at 3d anti declined distinctly at 5d; as compared with the fake group, it varied significantly （P 〈 0.01 ）. The number of positive cells of the PGXFD group was significantly lower than those of the other groups at 1d, 3d and 5d （P〈 0.01）, but not at 6h time point. Conclusion The activated Caspase- 3 increased significantly in the neuron around the hematoma after ICH. The expression of activated Caspase - 3 can be depressed through suppressing wind, removing phlegm, inducing resuscitation, promoting blood circulation and removing blood stasis, ect. The results suggest that these CTM remedys can prohibit neuronal apoptosis, especially Pinggan Xifeng decoction showed the best beneficial effect.
