摘  要:目的研究选择性环氧合酶(cyclooxygenase,COX)-2抑制剂尼美舒利和非COX抑制剂扑热息痛,对人卵巢癌细胞系SKOV3生长作用的影响。方法利用免疫组化法检测SKOV3细胞系中COX-2表达,以及两种药物对Ki-67在SKOV3细胞中表达的影响;透射电镜观察两种药物作用后,细胞形态变化及凋亡情况;流式细胞术检测两种药物作用后,对SKOV3细胞周期的影响。结果COX-2在SKOV3细胞中表达呈阳性,尼美舒利作用后的SKOV3细胞中,可发现Ki-67表达明显减少,扑热息痛对Ki-67表达无影响;透射电镜观察到细胞中凋亡小体的形成;流式细胞术发现,尼美舒利使SKOV3细胞停滞在G0/G1期,扑热息痛对细胞周期改变无影响。结论选择性COX-2抑制剂尼美舒利,可以作为卵巢癌的预防及治疗应用于临床。Objective The purpose of this study was to investigate the cellular effect of a selective COX-2 inhibitor (nimesulide) and aeetaminophen on the growth of SKOV3 cell line of human ovarian cancer in vitro. Methods SKOV3 cell line was cultured with nimesulide and aeetaminophen, immunohistochemical assessment was performed for alterations in expression of COX-2, Ki-67 enzyme. Cell cycle repartition was analysed by flow eytometry (FCM); The morphologie change and apoptosis of the cell were assessed under the electron microscopy (EM). Results COX-2 was presented positive in SKOV3 cell line and the reduction of Ki-67 were shown in immunohistoehemieal assay in the nimesulide. The cell apoptosis and necrosis were detected with nimesulide through EM. The cycle of cell was retention in G0/G1 by nimesulide. Conclusion Our suggest additional study for the use of COX-2 inhibitors, Nimesulide, as a strategy of chemoprevention for ovarian cancer,
