摘  要:目的:初步探讨DLC-1基因在结肠癌细胞sw480中的表达改变及其可能机制;方法:采用RT-PCR和Western blot分析结肠癌细胞CaCo2、sw480的DLC-1表达情况;应用去甲基化药物5—氮杂胞苷处理DLC-1基因阴性表达的细胞株,观察用药前后该细胞DLC-1基因的表达水平、细胞周期、细胞增殖力和侵袭力的影响。结果:CaCo2细胞高表达DLC-1 mRNA,而sw480则呈现表达缺失。经去甲基化药物5—氮杂胞苷处理sw480后,DLC-1 mRNA恢复表达,比较用药前后sw480细胞增殖力下降、细胞周期被阻滞在G2期、细胞侵袭力减弱。结论:DLC-1基因可影响结肠癌细胞sw480的增殖能力和侵袭力,并使结肠癌细胞sw480细胞周期阻滞于G2期,DNA甲基化可能是导致DLC-1在sw480细胞中的失活的一个重要原因。Objective: To investigate the expression of DLC-1 in the colorectal cancer cell line sw480. Methods: DLC-1 mRNA and protein expression was studied in the colorectal cancer cell line CaCo2 and sw480 using RT-PCR and Western blot. The colorectal cancer cell line nonexpressing DLC-1 was treated with 5-Aza-dC, and the change of DLC-1 expression can be detected which could effect the cell cycle, the activation in cell growth and the ability of invasion. Results: The expression of DLC-1 in the colorectal cancer cell line CaCo2 was detected frequently, but no in sw480. Treatment with 5-aza-2-deoxycyfidine (5-Aza-dC) induced DLC-1mRNA expression in the colorectal cancer cell line sw480, and the cell cycle was blocked in G2 cycle, the downregulation of activation in cell growth and the ability of invasion was detected, which identify the character of tumor suppressor gene of DLC-1. Conclusion: The cell cyde the activation in cell growth and the ability of invasion of the colorectal cancer cell line sw480 can be effected by DLC-1 gene, and the cell cycle was blocked in G2 cycle. In addition, the hypemethylaty within DNA maybe leads to the inactivation of DLC-1 of the colorectal cancer cell line sw480.
