摘  要:研究3种氮源(NaNO3,尿素,NH4NO3/尿素复合肥)对盐生杜氏藻(Dunaliella salina OUN07)生长和细胞生化组成的影响。结果表明,D.salina OUN07生长最适的是0.75mmol/L尿素,最大细胞密度为105×104cell/mL,对照组仅为59×104cell/mL;最高β-胡萝卜素含量110.6mg/g出现在0.125/0.125 mmol/L NH4NO3/尿素复合肥中,较低的氮盐有利于β-胡萝卜素的积累;1.0mmol/L尿素培养的D.salina OUN07叶绿素a含量最高(48.5mg/g),较高的氮盐有利于叶绿素a的合成;D.salina OUN07脂肪酸主要由16:0、18:1和18:2ω-6组成,其中0.75mmol/L尿素组脂肪酸含量最高;氮源对D.salina OUN07蛋白含量也有显著影响,NH4NO3/尿素组蛋白含量(33.61%)最高;建立了三种氮源吸收的动力学方程。Effects of three kinds of nitrogen sources (NaNO3, urea and NH4NO3/urea) on the growth and cell biochemical composition of Dunaliella salina were investigated. The results indicated that 0.75mmol/L urea was the optimum nitrogen source for the growth of D. salina OUN07 and the maximum cell density of 105 × 10^4cell/mL was detected in it, whereas the control group only had a cell density of 59 × 10^4 cell/mL. The maximum β- carotene content of 110.6 mg/g in D. salina OUN07 was detected in the medium with 0. 125/0. 125mmol/L NH4NO3/urea as nitrogen source, and low nitrogen was found to be benefit to β - carotene accumulation. The maximum content of chl a of 48.5 mg/g was found in the medium with 1. 0 mmol/L urea, and high nitrogen was found to be benefit to biosynthesis of chl a. The main fatty acids of D. salina OUN07 were composed of 16:0, 18:1 and 18:200-6 and the highest fatty acid content was detected in the medium with urea of 0.75 mmol/L as nitrogen source. The marked effect of different nitrogen sources on the protein contents was also detected and the highest protein content of 33.61% was recorded in the medium with NH4NO3/urea as nitrogen sources. The assimilated patterns of the three kinds of nitrogen sources by D. salina OUN07 were similar to each other and the assimilated equations of the three kinds of nitrogen sources by D. salina OUN07 were established.
