摘　　要：[目的]探讨细胞S相激酶相关蛋白2(skp2)和细胞周期素依赖性激酶抑制蛋白27(p27kip1)表达与胃癌发生的关系。[方法]用免疫组化SP法检测69例胃癌组织中skp2、p27kip1蛋白的表达,并与28例正常胃黏膜作对比;用RT-PCR方法随机检测其中3例胃癌组织中skp2的mRNA表达,并与其正常胃黏膜作对比。[结果]p27kip1蛋白阳性表达率在胃癌中为46.38%,而正常胃组织中为96.43%(P<0.01)。p27kip1蛋白在胃癌中的阳性表达率随肿瘤分化程度的降低、浸润深度的加深、淋巴结转移、病理分期进展而降低(P<0.05)。skp2mRNA在胃癌组织中的表达明显上调(P<0.01)。skp2蛋白阳性表达率在胃癌中为33.33%,而正常胃组织中为10.71%(P<0.05)。skp2蛋白阳性表达率随肿瘤的分化程度降低而升高(P<0.05)。skp2、p27kip1蛋白在胃癌中的表达呈负相关(P<0.01)。[结论]p27kip1、skp2蛋白检测有助于胃癌的临床诊断及判断预后。[Purpose] To explore the relationship between expressions of S-phase kinase-associated protein 2 （skp2）, cyclin dependent kinase inhibitor protein 27 （p27^kip1） and gastric carcinogenesis. [Methods] The protein expressions of p27^kip1 and skp2 were detected by immunohistochemistry SP method in 69 cases with gastric carcinoma, 28 cases with normal gastric mucous membrane were examined as control group. Three cases randomized from 69 cases with gastric carcinoma were examined by reverse transcription polymerase chain reaction method （RT-PCR） for the mRNA expression of skp2 and compared with their normal gastric mucous membranes as control. [Resultsl The positive expression rate of p27^kip1 protein was 46.38% in gastric carcinoma, while that was 96.43% in normal gastric tissue（P〈0.01）. The positive expression rate of p27^kip1 protein decreased with lower differentiation, deeper invasion, lymph node metastasis, advanced pathological stage （P〈0.05）. The expression of skp2 mRNA in gastric carcinoma was up-regulated obviously（P〈0.01）. The positive expression rate of skp2 protein was 33.33% in gastric carcinoma, while that was 10.71% in normal gastric tissue （P〈0.05）. The positive expression rate of skp2 increased with lower differentiation （P〈0.05）. There was a negative correlation between the expressions of skp2 and p27^kip1 in gastric carcinoma（P〈0.01）. [Conclusions] The examination of p27^kip1 and skp2 protein is helpful to clinical diagnosis and prognostic evaluation in gastric cancer.
