长期培养自体骨髓基质上清中白细胞介素3、白细胞介素6及肿瘤坏死因子和干扰素水平的变化

摘　　要：选择大连医科大学附属二院血液内科、大连市友谊医院血液科、铁路医院血液科、大连大学附属医院血液科初诊或复发的20例急性髓细胞白血病为实验对象,男11例,女9例,19～55岁,诊断均符合张之南的《血液病诊断及疗效标准》。长期培养白血病骨髓基质上清,取14,28,35d不同时间点的上清,应用ELISA法检测细胞因子水平。结果显示,培养14d的白细胞介素3含量低于28d及35d时间点(P<0.05),培养28d、35d其含量差异不显著(P>0.05);培养14d、28d的急性髓细胞白血病基质上清中白细胞介素6含量差异不显著(P>0.05),其含量均高于35d时间点(P<0.05);培养28d的肿瘤坏死因子α含量明显高于14d及35d时间点(P<0.05);14d与35d含量差异不显著;干扰素α含量随培养时间的延长而增高,各组间差异不显著。Totally 20 new or recurrent acute myeloid leukemia （AML） patients from Department of Hematology of Second Hospital Affiliated to Dalian Medical University, Department of Hematology of Dalian Friendship Hospital, Department of Hematology of Railway Hospital, Department of Hematology of Hospital Affiliated to Dalian University were selected, including 11 males and 9 females aged from 19-55 years. Their diagnosis was all based on The Standard of Hematology Diagnosis and Therapy, edited by 7hang. The supemate of AML stroma cells was cultured for a long term before supemate at different time points （14, 28, 35 days） were obtained. The cytokine content was detected by ELESA method. The results showed that the content of intedeukin （IL）-3 at day 14 was lower than that at days 28 and 35 （P 〈 0.05）. There was no significant difference between day 28 and day 35 （P 〉 0.05）. The content of IL-6 was similar at days 14 and 28 （P〉 0.05）, but they were higher than that at day 35（P 〈 0.05）. The content of tumor necrosis factor （TNF） a at day 28 was obviously higher than that at days 14 and 35 （P 〈 0.05）. There was no significant difference of the content between day 14 and day 35. The content of interferon a gradually increased during the time went on, which was not notable different at each group.
