摘  要:目的应用基因芯片筛选先天性肾盂输尿管连接部梗阻(UPJ)的相关基因。方法用一步法提取梗阻段输尿管及正常输尿管组织的总RNA,并过柱纯化,用包含21522条cDNA的基因芯片对梗阻段输尿管及正常输尿管的基因表达谱进行分析。结果肾盂输尿管连接部梗阻段输尿管与正常输尿管组织中差异表达的基因有709条,其中UPJ中下调的基因有535条,上调基因174条。表达差异有极显著意义的基因有16条,其中UPJ下调的基因有11条,上调基因5条。结论基因芯片可有效筛查出肾盂输尿管连接部梗阻发生的相关基因,包括细胞生长、发育、分化、信号传导、核酸转录、翻译、物质代谢及运输等多种基因,这表明在UPJ的发生发展中有多种类型的基因参与,非单一因素所致。Objective To study the gene expressions in the stenotic pelvi-ureteric junction by cDNA microarry. Methods Total RNA was isolated from the stenotic tissue of congenital pelvi-ureteric junction obstruction and normal ureters. RNA was purified with NucleoSpin RNA kit, converted to eDNA and hybridized using cDNA human microarry genes chip. Results The expressions of 709 genes were different between the stenotic tissue and normal ureter, of which 535 were down-regulated and 174 up-regulated in congenital pelvi-ureteric junction obstruction. Of these, 16 were significantly different, 11 of them down-regulated and 5 up-regulated. Conclusions cDNA microarry can be used to study possible genetic influence in the pathogenesis of congenital pelvi-ureteric junction obstruction.
