c—fos基因在衰老的成纤维细胞核染色质中结构状态的研究

摘　　要：为探讨衰老过程中促增殖基因在染色质结构存在的状态，用表皮生长因子（ＥＧＦ）刺激不同代龄的人胚肺成纤维细胞（２ＢＳ细胞），再用ＤＮａｓｅＩ超敏感方法对ｃ—ｆｏｓ基因进行检测，发现衰老２ＢＳ细胞ｃ—ｆｏｓ基因对ＤＮｅｓｅＩ的敏感性降低，并且不能通过ＥＧＦ的刺激而发生结构状态的改变，提示衰老细胞促增殖基因可能多处于“关闭”状态，与衰老细胞的增殖下降有关。o study the state of generation-promoting genes in chromatin structure of the senes-cent cells,c-fos genes were examined with DNase I hypersensitive assay after 2BS cells ofdifferent generation had been stimulated by EGF.The result shows that the sensitivity of c-fos genes in senescent 2BScells to Dnase I falls and their state of structure can not changewith the sitmulating of EGF.It is suggested that generation-promoting genes in senescentcells might often be in a'closed,state,which is related to the decrease of generation of thesenescent cells.
