摘  要:通过从油污土壤中分离筛选出的石油烃降解菌假单胞菌DS-Ⅲ,研究了在以烃为惟一碳源条件下该菌的生长特征、降解性与脱氢酶活性变化之间的关系。结果表明,DS-Ⅲ的脱氢酶活性滞后于其细胞生物量增长,适应后DS-Ⅲ的脱氢酶活性变化与柴油的降解趋势基本一致,DS-Ⅲ菌的对数生长期代时为4.05d,生长速率为0.25d-1。在培养温度32℃、pH6.5时,DS-Ⅲ的脱氢酶活性达到最高,在该条件下获得的米氏方程中,Km为159.16μg·L-1,Vmax为11.06μg·L-1·d-1。Pseudomonas DS-Ⅲ, the bacteria degrading petroleum hydrocarbon, were isolated from the oil-contaminated soil, while the diesel oil was used as the sole source of carbon and energy. The characteristic of growth and degradation ability of DS- Ⅲ, along with its dehydrogenase activity, were then investigated and compared. The increase of dehydrogenase activity of DS- Ⅲ showed a lag compared with the corresponding biomass. After the domestication period, the dehydrogenase activity changed similarly with that the diesel oil degraded. The genera- tion time and growth rate for DS-Ⅲ in logarithmic period reached 4.05 d and 0.25 d^-1, respectively. The optimum culture condition was found at pH 6.5 and 30 ℃, in which Michaelis equation can be acquired by calculation, with Michaelis constant(Km) being 159.16μ·L^-1, and Vmax11.06μg·L^-1·d^-1.
