摘  要:[目的]探讨Na+,K+-ATP酶β1亚基(ATP1B1)与肿瘤细胞增殖分化的关系,以及外源性ATP1B1对肿瘤细胞的影响。[方法]采用RT-PCR检测肿瘤细胞株、正常外周血单核细胞中ATP1B1的mRNA水平,以及经脂多糖(LPS)促进细胞增殖、二甲亚砜(DMSO)诱导肿瘤细胞分化后ATP1B1的mRNA水平变化,并观察通过转染技术增加肿瘤细胞中ATP1B1表达后细胞的生长情况。[结果]肿瘤细胞株中ATP1B1的mRNA水平明显高于正常单核细胞(P<0.05),促进细胞增殖后细胞中ATP1B1的mRNA水平增高(P<0.05),诱导分化后降低(P<0.05),导入外源性ATP1B1使肿瘤细胞增殖明显受抑(P<0.05)。[结论]Na+,K+-ATP酶β1亚基可以成为反映细胞功能状态特别是肿瘤细胞增殖代谢的重要指标,为肿瘤生物治疗的新策略提供重要的实验依据。[Purpose] To explore the relationship between Na^+,K^+-ATPase β1-Subunit(ATP1B1)and tumor cell in proliferation and differentiation,as well as the effect of ATP1B1 introduction.[Methods] The levels of ATP1B1 mRNA were detected by RT-PCR in both of tumor cell lines and normal peripheral blood monocyte,and the changing of ATP1B1 mRNA were analyzed in the proliferation-cells stimulated by LPS and the differentiation-cells induced by DMSO.Moreover,survival of cells transfected with ATP1B1 was observed.[Results]The expression of ATP1B1 mRNA in tumor cells transnificantly higher than that of normal monocyte (P〈0.05).ATP1B1 mRNA level was enhanced in tumor cell lines and monocytes by LPS stimulation (P〈0.05),while the level of ATP1B1 mRNA of these cells were decreased by DMSO-induced differentiation (P〈0.05).The growth of the tumor cells transfected with ATP1B1 was inhibited obviously (P〈0.05).[Conclusion]ATP1B1 can be the index of cell functional status,expecially tumor cell proliferation and differentiation.It will become an important experiment foundation for the biotherapy of tumor.
