摘  要:目的建立人子宫内膜癌SCID鼠模型,为子宫内膜癌的体内实验及临床研究提供合适的动物实验模型。方法分别选择低分化与高分化子宫内膜癌组织标本移植于SCID鼠皮下,原代移植瘤形成后进行鼠间传代,对移植瘤进行相关的生物学检测。结果成功建立3只原代人子宫内膜癌SCID鼠皮下移植瘤模型,其中低分化组织成瘤2只,中分化组织成瘤1只,原代移植成功率为37.5%,自第五代后,移植瘤的传代成功率为100.0%。病理组织学证实:连续传代的移植瘤仍保持原癌组织病理形态特点及人类肿瘤的特点。在子宫内膜癌SCID鼠皮下移植瘤模型中,100.0%移植瘤模型雌激素受体表达为阳性,33.3%孕激素受体表达为阳性,与有关文献报道一致。结论成功建立了人子宫内膜癌SCID鼠皮下移植瘤模型,移植瘤保持了原子宫内膜癌生物学特征,为人类子宫内膜癌的体内实验及临床研究提供了较理想的动物模型。Objective To establish tumor models of human carcinoma of endometrium in SCID mice,It will provide appropriate tumor models of human carcinoma of endometrium for experiental study. Methods Samples of human carcinoma of endometrium were subcutaneouly heterotransplanted to SCID mice. The initially transplanted tumor were transplanted to SCID mice for 6 passages in turn, the volumes of tumor were observed, pathology and the estrogen receptor and the progesterone peceptor (ER,PR) were inspected. Results Transplantable human carcinoma of endometrium were established successfully,two of poorly differentiated tissues and one of moderately differentiated tissue,the initial take rate was 37.5%, then it increased to 100.0% aftter the 5th passage. With the increasement of passage,the latent periods of tumor growth and the mean survival time were shortened obviously. The positive rate of ER,PR were 100.0% and 33.3%. HistologiCal resluts showed the transplanted tumors remianed the characteristics of original human carcinoma of endometrium. Conclusion The subcutaneouly transplantable human carcinoma of endometrium models had been established successfully,The transplanted tumors remianed the characteristics of original human carcinoma of endometrum. The models may be an ideal animal models for research of human carcinoma of endometrium.
