摘  要:目的探讨β-淀粉样蛋白25~35(amyloid-beta protein25~35,Aβ25~35)对体外培养血管平滑肌细胞(VSMC)的毒性作用。方法实验分对照组(生理盐水组)和Aβ25~35组(浓度分别为10、20、50、100μmol/L),分别培养1、3、10d,光镜下观察VSMC形态学改变;MTT法检测细胞活力;检测乳酸脱氢酶(LDH)活力。结果VSMC在浓度为20μmol/L的Aβ25~35孵育3d后即出现退行性改变,细胞存活率下降,LDH活力增高,其程度均呈Aβ25~35浓度、孵育时间依赖性。结论Aβ25~35可致VSMC形态学改变、存活率降低、LDH活力增高,有细胞毒性作用。Objective To investigate the toxicity of amyloid-beta protein 25-35 (Aβ25-35) on cultured vascular smooth muscle cells in vitro. Methods The control group was normal saline. In Aβ25-35 group, VSMC were incubated with Aβ25-35 at different concentrations (10,20,50,100 μmol/L) for ld, 3 d and 10 d. VSMC morphology was detected with .light microscope, vitality was measured by MTT method, and the lactate dehydrogenase concentration was tested. Results After incubated with 20 μmol/L Aβ25-35 for 3d, the survival rate was decreased and the lactate dehydrogenase level was increased and VSMC began to degenerate. The degree of cell degeneration, decreasing of survival rate and increasing of lac- tate dehydrogenase concentration all showed Aβ25-35 concentration and incubation time dependence. Conclusions The toxicity of Aβ25-35 on VSMC includes changing of cell morphology, decreasing survival rate and increasing lactate dehydrogenase level.
