摘 要:用流式细胞仪技术测定了K562/A02多药耐药细胞系和K562/S药物敏感细胞系细胞内柔红霉素(DNR)的含量,发现在2μmol/L的DNR孵育条件下,K562/A02细胞内DNR平均道峰值为72,K562/S细胞内DNR平均道峰值为112,细胞内DNR含量前者明显低于后者(P<0.01)。结果表明,流式细胞仪技术能较好地检测出细胞系对DNR细胞内蓄积的差异,提示此技术可鉴别多药耐药肿瘤细胞和敏感细胞,可用于临床多药耐药肿瘤患者的筛选。Abstract The daunorubicin content in K562/A02 multidrug resistant cell line and K562/S drug sensitive cell line was measured by flow cytometry . The results showed that daunorubicin mean peak channel number in K562/A02 and K562/S was 72 and 112 respectively when incubated with 2 umol/L daunorubicin. The quantity of daunorubicin in K562/A02 was much lower than that in K562/S (P<0.01). These results indicated that flow cytometry revealed the difference of the intracellular daunorubicin conant clearly between two cell lines. It also suggested that this technique was capable of differntiating multidrug resistant tumor cells from sensitive counterparts and is probably very useful in sorting patents with multidrug resistant tumor.Keywonts: multidrug resistance, flow cytometry , daunorubicin
