摘　　要：目的探讨HPLC-电化学检测法测定人血清中利培酮及9-羟利培酮浓度。方法采用美国A西-lentl100高效液相色谱仪联用ESA公司库仑阵列电化学检测器。四个通道的电极依次设置为500，700，850，950mV，用shim-packVP-ODS色谱柱（4．6mm×150mm，5μm），甲醇-80mmol·L^-1乙酸钠（60：40）为流动相，流速为1mL·min^-1，去甲咪嗪为内标，甲苯为萃取溶剂。结果利培酮在0．625～60ng·mL^-1范围内线性良好；r=0．9967，日内RSD≤8．59％，日间RSD≤9．45％，相对回收率在93．4％-102．4％。9-羟利培酮在1．25～120ng·mL^-1范围内线性良好；r=0．9985，日内RSD≤8．47％，日间RSD≤8．54％。相对回收率在99．2％～106．9％。结论该方法灵敏、准确、快速、可用于利培酮临床血药浓度监测和药动学研究。Objective To explore an HPLC method for quantitative determination of risperidone and 9 - hydroxyrisperidone in human serum. Methods Their concentration were determined by HPLC with a coulometric array detector （CAD）. The CAD has 4 electrochemical detector cell. The cell potentials were maintained at 500,700,850,950mV. A shim - pack VP - ODS chromatographic column（4. 6mm × 150mm,5μm） was used, mobile phase was consisted of MeOH - 80 mmol · L^-1 NaAc（60： 40） ,flow rate was 1mL · min^-1 ,the internal standard was desipramine,serum sample were extracted with toluene. Results A linear range of risperidone was 0. 625 - 60ng · mL^ - 1, （ r = 0. 9967）, the intra - and inter - run precision were lower than 8.59% and 9. 45%, the method recovery was 93.4% - 102. 4%. A linear range of 9 - hydrexyrisperidone was 1.25 - 120ng · mL^- 1, （ r = 0. 9985 ）. The intra - and inter - run precision were lower than 8.47% - 8.54% ,the method recovery was 99. 2% - 106. 9%. Conclusion The method is sensitive, simple and accurate, and can be used for clinical monitoring and its pharmacokinetic studies.
