摘  要:目的 观察不同剂量戊四氮(PTZ)致SD大鼠慢性点燃癫(癎)模型的效果.方法 采用PTZ腹腔注射制作SD大鼠慢性点燃模型,并将其分为对照组、癫(癎)Ⅰ组[PTZ,30 mg/(kg·d)]、癫(癎)Ⅱ组[PTZ,35 mg/(kg·d)]、癫(癎)Ⅲ组[PTZ,40 mg/(kg·d)];观察发作情况;分别计数雌雄鼠发作只数并对发作后大鼠脑组织行尼氏染色.结果 Ⅰ,Ⅱ组之间潜伏期无显著差异;Ⅲ组与Ⅰ,Ⅱ组之间潜伏期差异有统计学意义(P<0.01),Ⅲ组潜伏期最短,但死亡率高;雌雄鼠在各剂量条件下未见显著差异;癫(癎)组比对照组明显的组织增生,各剂量癫(癎)组织间无显著差异.结论 在该实验中,PTZ腹腔注射致SD鼠点燃慢性癫寮模型的最适剂量是30~35 mg/(kg·d).Objective To observe the effectiveness in triggering epleptic rat model by intraperitoneal injection of different doses of pentetrazole (PTZ).Methods Spraque- Dawley rats were divided randomly into four groups:control group, epilepsy Group Ⅰ (PTZ,30 mg/ (kg·d) ,epilepsy Group Ⅱ (PTZ,35 mg/kg·d) and epilepsy Group Ⅲ (PTZ,40 mg/kg·d).The epileptic rat model was developed by daily intraperitoneal injection of PTZ. The seizures in the rats were observed. Results There was no significant difference in latent phase between Group Ⅰ and Group Ⅱ, but there was between Group Ⅲ and Groups Ⅰ , Ⅱ Group Ⅲ showed the shortest latent phase but also the highest death rate;There were no significant differences between male and female rats in all three epilepsy groups. The epilepsy groups had more significant hyperblastosis than the control group.There were no significant differences between the three epilepsy groups. Conclusions In this experiment,the optimal dose of PTZ for developing epilepsy rat model is 30- 35 mg/(kg·d).
