真核表达Survivin基因对卵巢癌细胞紫杉醇敏感性的影响

摘　　要：目的探讨Survivin基因对卵巢癌细胞紫杉醇(Taxol)敏感性的影响。方法构建Survivin正义全长真核表达质粒(pcDNA3.1-Survivin),经脂质体包裹转染人卵巢癌细胞A2780,以转染pcDNA3.1空载体的A2780细胞为对照。RT-PCR和蛋白质免疫印迹(Western blot)法检测Survivin mRNA和蛋白质的表达;MTT比色法检测紫杉醇对两组细胞存活率的影响;流式细胞仪(FACS)检测细胞凋亡率。结果①RT-PCR和Western blot检测提示转染pcD-NA3.1-Survivin组中Survivin mRNA和蛋白质表达明显高于空载体组。②MTT比色法和FACS检测提示转染pcD-NA3.1-Survivin组细胞存活率明显高于空载体组,细胞凋亡率明显低于空载体组,差异均有显著性意义(P<0.05)。pcDNA3.1-Survivin转染后的A2780细胞对紫杉醇的敏感性明显降低。结论卵巢癌对紫杉醇的耐药性可能与Sur-vivin基因表达有关。Objective To investigate the effect of Survivin expression on sensitivity of ovarian cancer cells to Taxol. Methods To construct Survivin eukaryotic expression plasmid, pcDNA3. 1-Survivin （full-length Survivin cDNA）, we transfeeted A2780 cells with pcDNA3.1-Survivin by Lipofectamine. The expression of Survivin mRNA and protein was detected by reverse transcription-polymerase chain reaction （RT-PCR） and Western blot, respectively. Cells were treated with Taxol and analyzed for viable rate and apoptotic rate by MTT assay and flow cytometry （FACS）. Results ① The level of Survivin mRNA and protein of cells transfected with pcDNA3. 1-Survivin was significantly higher than control detected by RT-PCR and Western blot. ②The viable rate of cells transfected with pcDNA3.1-Survivin was obviously higher than control and the apoptotic rate was significantly lower than control detected by MTT assay and FACS, respectively. There was significant difference between two groups. The sensitivity of A2780 cells transfected with pcDNA3. 1-Survivin to Taxol was signifieantly decreased. Conclusion The drug resistance of ovarian cancer cells to Taxol may be related to expression of Survivin gene,
