摘  要:目的为开发利用平白术的药用价值提供理论依据,分别提取分离平白术中有效成分多糖和氨基酸,并分别测定平白术多糖和氨基酸的含量。方法采用L9(34)实验因素设计实验,苯酚-硫酸法测定白术多糖的含量。以茚三酮为衍生试剂,采用柱后衍生法测定平白术中氨基酸含量。结果平白术多糖的含量为15.3%。平白术中至少含有17种氨基酸,总氨基酸含量为73.73 mg.g-1。结论方法简单、有效、快速、准确、灵敏,适合于分析测定白术中多糖和氨基酸的含量。Objective To extract polysaccharide and amino acid and determine their contents in order to offer the basis for using Atractylodes macrocephala Koidz from Pingjiang. Methods Using L9 (34) orthogonal test, Phenol-sulfuric method was used to test polysaccharide content in Atractylodes macrocephala and to assay amino acid using method of Ninhydrin post - column derivation. Results The analysis ofAtractylodes macrocephala Koidz showed that the content of polysaccharide was 15.3% and there were at least 17 kinds of amino acids,the total content was 73.73 mg·g^-1. Conclusion The established method is quite suitable for identification and quantification of polysaccharide and amino acids in Atractylodes macrocephala Koidz. It is simple ,effective, rapid, accurate and sensitive.
