摘 要:目的探讨热疗联合放疗对小鼠S180肿瘤模型细胞凋亡及血管再生的影响。方法实验用昆明小鼠60只,右后肢股部皮下接种S180肿瘤细胞株,当肿瘤生长至1cm3时分组进行局部热疗、放疗和热放疗。然后用RT-PCR检测Bcl-2 mRNA,免疫组化方法检测调亡细胞,血管内皮生长因子(VEGF)蛋白,以微血管密度(MVD)作为定量检测指标。结果热疗、放疗、热放疗组细胞凋亡指数较对照组增加(P<0.01),以热放疗组增加最为显著(P<0.01)。热疗、放疗、热放疗组Bcl-2基因表达较对照组减弱(P<0.01),以热放疗组减弱最为显著(P<0.01)。热疗组VEGF蛋白表达和MVD较对照组降低(P<0.01),放疗组VEGF蛋白表达和MVD较对照组升高(P<0.01),热放疗组VEGF蛋白表达和MVD较放疗组降低(P<0.01)。结论热疗可以下调Bcl-2基因,诱导S180株细胞凋亡,抑制肿瘤细胞VEGF的产生,使MVD下降。与放疗共同作用于肿瘤细胞,可增加肿瘤细胞放射敏感性,对肿瘤有显著的治疗作用。Objective To study the effects of hyperthennia combined radiation on apoptosis and angiogenesis of S180 tumor cells.Methods S180 tumor cells were injected subcutaneously into 60 rats in back-right leg, which were randomly assigned into 4 groups. When the tumors volume reached, 1cm^3 , hyperthennia, radiation, hyperthennia combined radiation were given respectively. Then Bcl-2 mRNA was measured by RT-PCR;Apoptosis, VEGF and MVD were measured by immunohistochemical staining. Results Compared with the control group, the apoptosis fraction of hyperthennia(H), radiation(R), hyperthennia combined radiation(HR) group were increased( P 〈 0.01) ,the Bcl-2 mRNA were decrease( P 〈0.01 ),the VEGF and MVD of H were decreased(P 〈0.01),that of R were increased( P 〈 0.01).The VEGF and MVD of HR were decreased compared R( P 〈 0.01 ). Conclusion Hyperthennia can depress Bcl-2 mRNA, induce S180 cell apoptosis, and restrain tumor cells produce VEGF protein. Hyperthermia combined radiation can improve the tumor cell sensitivity to irradation and become a better therapy to tumor patients.
