摘  要:目的研究薏苡仁油对人乳腺癌细胞系MCF-7的凋亡诱导作用及机理。方法在体外设定不同浓度的薏苡仁油处理MCF-7细胞系的实验组,细胞培养24 h后,用MTT法测定其细胞活力;碘化丙啶(PI)染色后流式细胞仪检测其凋亡情况;罗丹明123(Rodamine 123)标记后于酶标仪530 nm处测定其荧光强度值以检测线粒体膜电位情况。结果与对照组相比,随薏苡仁油浓度的上升,MCF-7细胞活力和增殖呈剂量依赖型的下降;流式细胞仪检测PI染色显示,各处理组的MCF-7细胞凋亡率随作用浓度的上升而增强,最高浓度组尤为明显;在高浓度组,罗丹明123荧光强度明显降低。这些结果表明薏苡仁油能抑制MCF7细胞的活力和增殖;诱导其凋亡;罗丹明荧光强度的减少,说明MCF-7线粒体膜电位有倒塌和去极化的情况发生,提示MCF-7细胞的凋亡与线粒体有关。结论薏苡仁油可明显促进MCF-7细胞的凋亡,其凋亡可能与线粒体破坏有关。In this research we studied the effects of coix lacrymajobi L varma-yuen lipid on MCF-7 cells, which were obtained from human breast adenocarcinoma. After being treated with different concentrations for 24 hr, the viability of MCF-7 cells was detected by MTT; apoptosis was determined by FCM after being stained with propidium iodine (PI), and the membrane potential of mitochondria was detected by fluorescence intensity after being stained with Rodamine 123. The results showed that after being co-cultured for 24 h, the viability of MCF-7 cells had a down-regulated tendency with the increase of concentrations; the apoptosis percentage in high dose class was more significant than that in lower-dose treated classes; the mitochondrial membrane potential collapsed after being treated with coix lacrymajobi L varma-yuen lipid for 24 h, which was especially evident in high-dose stimulated columes.
