卡介苗Ag85B对小鼠膀胱癌治疗作用及机制的研究

摘　　要：目的探讨卡介苗(BCG)Ag85B蛋白对小鼠膀胱癌的抑癌效应及免疫机制。方法采用T739小鼠皮下膀胱癌模型,肿瘤局部皮下注射BCG Ag85B蛋白,并用BCG和生理盐水作对照,观察各组的肿瘤重量和存活期,利用ELISA方法检测各组的3种主要细胞因子的水平,电镜和光镜观察各组肿瘤细胞的坏死和凋亡情况,利用流式细胞技术检测各组肿瘤细胞凋亡和细胞增殖周期。结果BCG Ag85B组小鼠肿瘤瘤重显著低于其它两组,其差异有统计学意义(P<0.05),该组小鼠存活天数显著长于其它2组,P<0.05;该组小鼠血清中白细胞介素-2、肿瘤坏死因子-α和干扰素-γ的水平明显高于其它2组,P<0.05;电镜和光镜下观察Ag85B组坏死明显增加。细胞增殖指数下降,其差异有统计学意义(P<0.05)。结论BCG Ag85B蛋白对T739小鼠膀胱癌具有抑制作用,并能延长荷瘤鼠的生存期,不论从细胞因子水平和光镜与电镜观察坏死和凋亡情况,还是从细胞凋亡和增殖情况来看Ag85B蛋白均优于卡介苗。Objective To research into the therapeutic effect and immunological mechanism of Ag85B to BTT739 murine BTCC. Methods We establish routine hypodermical transplantable BTCC model, inject hypodermically with the same volume of Normal saline, BCG and Ag85B near the carcinoma in each group, observe the weight of tumor and the survival days, detect the level of IL-2, TNF-α and IFN-γof mouse serum in each group, observe necrosis and apoptosis of tumor cells with light microscope and transmission electron microscope, and detect proliferation index and proportion data of apoptosis of mouse tumor cells by FCM. Results The mean tumor weight of the Ag85B group is much lower than that of other two groups with statistical difference; the survival period in this group is longer, and the difference between the three groups is statistically significant; the level of IL-2, TNF-α and IFN-γin this group is much higher than the rest with statistical difference. Tumor necrosis of Ag85B group is much more serious than the others with light microscope and the proliferation index of Ag85B group was the lowest statistically. Conclusion From the above we can conclude that the protein of BCG Ag85B can suppress the tumor growth of BTT739 and the therapeutic effect and the immune reaction is better than BCG.
