摘  要:目的研究吉非罗齐对高脂饮食喂养的Wister大鼠肝细胞脂肪变性的影响。方法(1)72只Wister雄性大鼠随机分为基础饲料组(C)、高脂饲料组(F)和高脂加吉非罗齐组(FG),C、F、FG组各含1、2、4、8周4个时相点,每组6只动物。(2)动态观察各组:①动物肝重指数、丙二醛含量的变化;②光镜观察肝脏病理变化;③分析大鼠肝脏PPARαmRNA和蛋白水平。结果(1)动物肝重指数的变化情况:8周后,FG>F>C,各组之间差异有统计学意义(P<0.05);(2)FG组PPARα的mRNA和蛋白表达显著升高,明显高于F组和C组;(3)肝脏内MDA含量在F组明显高于FG和C组(P<0.05),随时间延长,F组MDA逐渐增加(P<0.05);FG组随时间延长,其含量逐渐降低(P<0.05),8周时与正常相近(P>0.05)。结论(1)使用吉非罗齐增强大鼠PPARα的表达不会加重脂质过氧化作用对肝脏的组织形态和功能的损伤;(2)使用吉非罗齐增强大鼠PPARα的表达,对大鼠肝脏脂肪变有一定保护作用。Objective To investigate the effect of gemfibrozil on the hepatic fatty denaturalization of Wistar rats fed by high fat diet. Methods (1)Wister male rats were randomly divided into basic diet-treated group (C group), high fat diet-treated group (F group) and high-fat diet and gemfibrozil-treated group (FG group). Meanwhile,each of the groups was divided into 4 subgroups (1,2,4,8 week) respectively. Each subgroup had 6 rats. (2)The following parameters were observed dynamically in each group..① changes of animal liver weight index and the level of malondialdehyde (MDA) in liver tissue, ②pathological changes of liver observed by optical microscopy,③mRNA and protein expression of peroxisome proliferator activated receptorα (PPARα) in rat liver. Results (1)After 8 weeks, animals' liver weight index in H group was significantly higher than that in other groups(P〈0.05), and no obvious difference in other subgroups was observed (P〉0.05) ;for liver weight index, the order was FG group 〉 F group 〉C group, and the differences in every two groups were distinctive (P〈0.05). (2)The level of malondialdehyde (MDA) in liver tissue in F group was significantly higher than that in FG group and C group, respectively. And with time passing by, they increased gradually and remarkably (P〈0.05). In FG group, they decreased gradually (P〈0.05) and restored to normal at 8 week. (3)The mRNA and protein expression of PPARα in FG group increased significantly as time passing by and significantly higher than that in F and C group, respectively (P〈0.05). Conclusion (1) Gemfibrozil could enhance PPARα expression, which has no obvious effect on hepatocytes' morphology and function. (2) Enhanced PPARα expression has protective role to prevent hepatic fatty denaturalization.
