摘  要:目的以新西兰大白兔为动物模型评估Vitreon作为眼内填充物的安全性。方法24只(24眼)兔随机分为4组:玻璃体切割组(8只)、晶状体玻璃体切割组(8只)、对照玻璃体切割组(4只)及对照晶状体玻璃体切割组(4只),手术采用扁平部玻璃体切割术,经玻璃体切割及晶状体玻璃体切割分别填充Vitreon,术后观察1周、2周、4周、8周后,行房角光镜及视网膜光镜、电镜检查。结果光镜示Vitreon填充眼术后2周出现房角及虹膜基质慢性炎症,术后2个月房角基本消失。下方视网膜术后1周神经纤维层可见大量空泡,术后1个月见视网膜表面机化膜及吞噬细胞;2个月视网膜变薄,以核层及光感细胞层为著。电镜示术后2周色素上皮细胞吞噬外节膜盘;2个月外节膜盘大部溶解、破碎。结论Vitreon注入玻璃体腔后短期内出现视网膜病理改变,不宜作为眼内长期填充物。Objective To evaluate the safety of Vitreon used as intraocular implants in the model of New-Zealand rabbit. Methods Twenty-four rabbits (24 eyes) were random divided into four groups: lensectomy and vitrectomy group(8 eyes ), vitrectomy group ( 8 eyes), control lensectomy and vitrectomy group(4 eyes),control vitrectomy group( 4 eyes). The eyes were filled with Vitreon in vitrectomy,the retina and chamber angle were evaluated by light and/or electron microscopy at 1 week, 2 weeks, 4 weeks, and 8 weeks after operation. Results Histologically chronic inflammatory reactions in the inferior trabecular meshwork and iris stroma,and closure of the chamber angle were observed at 2 months after operation. Vacuoles in the nerve fiber layer in the inferior retina were observed after 1 week. Membrane tissue and macrophages were found after 1 month. Thinning of all layers of retina was observed, specially for the nulear and photoreceptor layer in the inferior retina after 2 months. After 2 weeks after operation, the electron microscopy examination showed that the retinal pigment epithelial cells engulfed the membranous disc of outer segments; The membranous disc of outer segment were broken or disappeared in the inferior retina at 2 months after operation. Conclusion Vitreon is not suitable as the long-term intraocular implants for the damages to the retina.
