摘  要:目的探讨调节性T细胞(Treg细胞)的异常在大鼠溃疡性结肠炎动物模型发病机制中的作用。方法40只Wistar大鼠分为正常对照组和溃疡性结肠炎模型组,应用三硝基苯磺酸/乙醇法建立溃疡性结肠炎的大鼠模型,流式细胞仪分析其外周血单个核细胞(PBMC)和肠系膜淋巴结(MLN)中的Treg细胞和CD4+T细胞的比例,实时荧光PCR测定Foxp3mRNA的含量。结果溃疡性结肠炎模型组大鼠PBMC中Treg细胞的比例明显高于正常(P<0.05),CD4+T细胞的比例明显下降(P<0.05),Foxp3 mRNA的含量明显增高(P<0.05);而MLN中Treg细胞的比例明显低于正常(P<0.05),CD4+T细胞的比例明显增高(P<0.05),Foxp3 mRNA的含量明显下降(P<0.05)。结论Treg细胞的异常在溃疡性结肠炎的发病机制中起重要作用。Objective To investigate the role of Treg ceils in the pathogenesis of ulcerative colitis in rats. Methods Studies were conducted in 40 Wistar rats which were divided into model group and control group. After the establishment of the model of ulcerative colitis by using trinitrobenzene sulfonic acid(TNBS)/ ethanol method, the proportion of Treg ceils and and CD4^+ T cells were detected by using flow cytometry analysis, at the same time, the expression of Foxp3 mRNA was measured in peripheral blood monocyte(PBMC) and mesenteric lymph node(MLN). Results 1. The proportion of Treg cells increased and the proportion of CD4^+ T cells decreased significantly in model group compared with that in control group in PBMC (P〈0.05) ; while The proportion of Treg ceils decreased and the proportion of CD4^+ T ceils increased sharply in model group compared with that in control group in MLN(P〈0. 05). 2. The expression of Foxp3 mRNA increased markedly in model group compared with that in control group in PBMC(P〈0.05), while decreased sharply in MLN(P〈 0.05). Conclusion The abnormal proportion of Treg cells plays an important role in the pathogenesis of ulcerative colitis.
