摘 要:目的探讨谷氨酸及谷氨酸载体在帕金森病(PD)发病机制中的作用。方法首先提取PD患者的血小板线粒体,然后分别先后用谷氨酸和草酰乙酸进一步装载这些线粒体,加入^(14)C-谷氨酸后启动谷氨酸载体的转运,同时测定该载体的活性,并观察多巴胺等药物对该载体活性的影响。结果PD患者的血小板计数无明显变化,但谷氨酸载体的活性明显降低,与对照组相比差异有显著性意义(P<0.05)金刚烷胺(Ama)可明显提高该载体的活件(与加药前相比,P<0.05),而多巴胺、左旋多巴和安坦对其活性无明显影响(与加药前相比,P>0.05)。结论谷氨酸以及谷氨酸载体参与了PD的发病机制,Ama可能通过影响谷氨酸载体的活性发挥治疗作用。Objective To investigate the roles of glutamate and glutamate carriers in the underlying mechanisms ofParkinson's disease (PD). Methods The platelet mitochondria of patients with PD were extracted, to load the mitochondria with glutamate and oxaloacetic acid respectively, then to trigger the transporting of the glutamate carrier by adding the ^14C-glutamate and determine the activities of the carriers, and finally to observe the changes of the activity of the carriers in the presences of dopamine, L-dopa, trihexiphenidyl and amantadine. Results There was no significant change in the platelet counts in the PD patients, but the activity of glutamate carrier obviously decreased in them as compared with the control group (P〈0.05). The amantadine could increase the activity (P〈0.05 vs non-drug treatment group). Other drugs used in our study showed no effects (P〉0.05 vs non-drug treatment group). Conclusion The glutamate and its carriers do play a role in the pathogenesis of Parkinson's disease, and the therapeutic efficacy of amantadine could be partly relevant to its effect on glutamate carrier.
