摘 要:目的:观察血管性痴呆小鼠海马结构生长抑素(SS)和胆囊收缩素(CCK)阳性神经元的数量变化,探讨血管性痴呆的发生机制。方法:复制小鼠血管性痴呆模型,利用Y-迷宫检测血管性痴呆模型小鼠学习记忆能力,采用ABC免疫组织化学方法,研究血管性痴呆小鼠与正常小鼠海马结构SS和CCK阳性神经元数量的变化。结果:血管性痴呆小鼠比正常小鼠Y-迷宫学习记忆训练次数明显增多,差异有统计学意义;海马结构CA1区、CA3区及齿状回(DG)区SS和CCK阳性神经元的数量明显减少,差异有统计学意义。结论:血管性痴呆的发生可能与海马结构CA1区、CA3区及齿状回(DG)区SS和CCK阳性神经元的数量明显减少有关。Objective: To observe the changes of the expression level of somatostatin (SS) and cholecystokinin (CCK) in CA1, CA3 and DG of hippocampal formation in mice with vascular dementia and investigate the mechanism of vascular dementia formation. Methods: Model of vascular dementia mice was duplicated; ABC immunohistochemistry were used to investigate the changes of SS and CCK in different groups and Y-maze test was used to observe the changes in learning and memorizing abilities. Results: Compared with the control group, the learning and memorizing abilities of the vascular dementia group were significantly decreased; the numbers of SS positive neurons and CCK positive neurons in the hippocampal formation CA1, CA3 and DG were significantly decreased (P〈0.05), Conclusion: The pathogenesis of the vascular dementia is probably related to the decrease of the number of SS positive neurons and CCK positive neurons in CA1, CA3 and DG of the hippocampal formation.
