摘 要:目的探讨柴苓归芪汤对脂质过氧化损伤的保护作用。方法SD大鼠36只随机分为模型组、辛伐他汀组、柴苓归芪组和正常组。模型组、辛伐他汀组和柴苓归芪组采用尾静脉一次性注射阿霉素5mg/kg复制肾病综合征大鼠模型,正常组注射等量0.9%氯化钠注射液。选模1周后开始灌胃,辛伐他汀组给予辛伐他汀1mg/(kg.d),柴苓归芪组给予柴苓归芪汤煎剂27g(kg.d),蚓激酶每日给予1次(2.5mg/100g,1500U/mg),共用药5周。正常组和模型组灌服等量蒸馏水。分别用黄嘌呤氧化酶法和硫代巴比妥法检测血清及肾组织超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量。结果模型组血清及肾组织中SOD活性均较正常组明显降低(P<0.05,P<0.01),而MDA含量却较正常组升高(P<0.05,P<0.01),提示阿霉素肾病(AN)大鼠清除氧自由基能力降低;柴苓归芪组血清和肾组织SOD活性明显升高P<0.05,P<0.01),而MDA含量明显降低(P<0.05,P<0.01),提示柴苓归芪汤不仅能提高全身氧自由基清除能力,而且能改善肾脏局部脂质过氧化状态。结论柴苓归芪汤能够有效的增强肾病综合征大鼠血清及肾组织SOD活性,降低血清及肾组织MDA含量,从而提高机体抗氧化能力,减轻脂质过氧化损伤。Objective To study the effect of Chailingguiqi decoction on injury of lipid peroxidation. Methods 36 rat model of nephrotic syndrome were duplicated by injection of Adriamycin. Rats were divided into model greup,Simvastatin group, Chailingguiqi decoction group and control group. Rats in model sroup and control group were tabken by distilled nater. Rats in Simvasfatin group and Chailimgguiqi group administrated separately Simvastatin (1 mg/(kg·d) )and Chailingguiqi decoction. The contents of SOD and MDA in serum and tissue were detected by method of xanthinoxidase and thio-barbitone. Results SOD activity in serum and tissue in model group were decreased compared with those in control group. The content of MDA in serum and tissue in model group were increased compared with those in control group. The results showed that Adriamycin-indnced nephropathy rat loss ability of removing oxygen free radical. SOD activity in serum and tissue in Chailingguiqi decoction group were increased. The content of MDA in serum and tissue in Chailingguiqi decoction group were decreased. Conclusion Chailingguiqi decoction can effectively increase SOD activity in serum and tissue and decrease content of MDA in serum and tissue, then elevate ability of antioxygen free radical.
