摘 要:目的研究T淋巴瘤侵袭转移诱导因子1(Tlymphoma invasion and metastasis inducingfactor 1,Tiam1)与Ras相关的C3肉毒素底物1(ras-related C3 botulinumtoxin substrate,Rac 1)信号转导及检测其在胃癌组织、癌旁组织、胃良性病变组织中的量值水平,进一步探讨其在胃癌转移中的作用。方法分别采用实时荧光定量聚合酶链反应(real-time fluorescence quantitive PCR,FQ-PCR)方法、RT-PCR(reverse transcription-polymerase chain reaction)方法检测66例胃癌、癌旁组织及11例胃良性病变中Tiam1 mRNA、Rac1 mRNA的表达量值水平。结果66例胃癌患者的癌和癌旁组织中Tiam1 mRNA表达阳性率分别为92.42%、19.60%,11例胃良性病变组织检测到1例Tiam1 mRNA表达,三者差异有统计学意义(χ2=78.96,P<0.01);胃癌和癌旁组织中Rac1 mRNA表达阳性率分别为81.82%、27.27%,两组差异显著(χ2=39.60,P<0.01),11例胃良性病变中无Rac1 mRNA的表达。胃癌组织中Tiam1 mRNA表达量(Md=4.92×105)高于癌旁组织(Md=0)。胃癌组织中Rac1 mRNA平均表达水平(Md=1.75,Q75=1.986)明显高于癌旁组织(Md=0.195,Q75=0.744),两者差异均有显著性(P<0.01);Tiam1、Rac1在不同临床分期、分化程度和胃癌转移的标本中的阳性表达率比较,差异有显著性意义(P<0.05);Tiam1、Rac1表达量值水平呈正相关(r=0.854,P<0.01)。结论Tiam1、Rac1 mRNA的高表达与胃癌的发生、转移密切相关,Tiam1、Rac1协同促进胃癌转移,为胃癌基因治疗提供了新的靶点。Objective To detect the expression quantity and To investigate signal transduction of ras-rehted C3 botulinum toxin substrate, Rac1 and T lymphoma invasion and metastasis inducing factor 1 ,Tiam 1 in gastric cancer tissues.tlssues around the cancer andgastric benign lesion tissues,and To investigate its effect in gastric cancer metastatic.Methods Respectivdy Tiam1 and Rac1 expression by real-time fluorescence quantitive PCR(FQ -PCR) and reverse transcription-polymerase chain reaction (RT-PCR) in 66 cases of gastric cancer tissues, tissues around the cancer and 11 cases of gastric benign lesion tissues.Results Positive rate of Tiam1 mRNA expression in gastric cancer tissues, tissues around the cancer is 92.42%. 19.6%, 1 respectively, only one case express Tiam1mRNA in 11 cases gastric benign lesion tissues,there was significant difference( P 〈 0.05) ;Positive rate of Rac1 mRNA expression in gastric cancer tissues and tissues around the cancer is 81.82% and 27.27 %, respectively, and there was significant differenee( Х^2 = 39.6, P = 0.001) ; There was no Rac1 mRNA expression in 11 cases of gastric benign lesion tissues. Expression of Tiam1 mRNA in gastric cancer tissues is higher than it in tissues around gastric cancer. Expression of Rac1 mRNA in gastric cancer tissues is obviously higher than it in tissue around the cancer, there was significant difference( P 〈 0.01). There was significant difference in the expression of Tiam1 and Rac1 among the different clinical stages,differentiated degree and tumor metastases in human gastric cancer tissues(P 〈0.05). Conclusion Hypsi-expression of Tiam1 and Rac1 mRNA was rahted with development and metachoresis in intestinal tract tumor. Tiam1 and Rac1 promote mutually gastric cancer metastatic and it might provid with new target for gene therapy in gastric cancer.
