离体培养下氮素浓度对小麦籽粒HMW-GS表达动态和含量的影响

摘  要:利用小麦穗离体培养技术和SDS-PAGE技术,研究了不同氮素浓度处理下小麦籽粒高分子量谷蛋白亚基(HMW-GS)的表达和积累规律。研究显示:氮素浓度不影响HMW-GS组成和表达顺序,各处理下HMW-GS的表达顺序均依次为亚基8、亚基7、亚基2和亚基12。但各亚基的表达时间与含量明显受氮素浓度影响:低氮浓度处理与高氮浓度处理相比,亚基8、亚基7、亚基2和亚基12的表达时间分别从花后121、5和18 d提前到花后6和9 d;HMW-GS的含量均与氮素浓度呈正相关,特别是亚基2不仅与氮素浓度呈极显著正相关,且影响亚基12对氮素的响应。Development and accumulation law of HMW- GS was studied in different nitrogen content by SDS - PAGE technology in wheat in vitro. The results showed that nitrogen concentration didn't affect the composition and the expression order of HMW - GS. The expression order of HMW - GS was subunit 8, subunit 7, subunit 2 and subunit 12 in turn in different nitrogen concentration, but the expression time and content of each subunit were affected by nitrogen concentration obviously. The expression time of subunit 8, subunit 7, subunit 2 and subunit 12 was advanced ahead of time after flower 6 d and 9 d from the time after flower 12 d, 15 d, 18 d individually in higher nitrogen content. A significant positive correlation was found between the content of HMW - GS and the different nitrogen concentration, especially the subunit 2. The subunit 2 was not only extremely remarkable positive correlation with the nitrogen concentration, but also affect the nitrogen response of the subunit 12.
