摘 要:目的:探讨大鼠大脑海马立体定向注射β-淀粉样蛋白1-40对Bax和Bcl-2mRNA表达的影响及通络救脑口服液对其干预作用。方法:140只Sprague-Dawley(SD)大鼠随机分为空白对照组、假手术组、阴性对照组、阳性对照组、实验组(12、24、48mg.kg-1.d-1)共7组。采用大脑海马立体定向注射β-淀粉样蛋白1-40诱导Alzheimer's病动物模型后,药物干预4周,第5周应用实时定量PCR法检测Bax和Bcl-2mRNA表达。结果:与假手术组比较,模型组Bax2-ΔΔCt明显升高,而Bcl-22-ΔΔCt明显降低,差异具有统计学意义(P<0.05),表明模型组Bax mRNA表达上调,而Bcl-2mRNA表达下调。与模型组比较,盐酸多萘哌齐组、通络救脑口服液各剂量组Bax2-ΔΔCt显著降低,表明Bax mRNA表达下调;而盐酸多萘哌齐组、通络救脑口服液各剂量组Bcl-22-ΔΔCt显著升高,表明Bcl-2mRNA表达上调。结论:大脑海马注射β-淀粉样蛋白1-40后大鼠海马组织Bax mRNA表达水平明显增高,Bcl-2表达明显降低,而通络救脑口服液可以抑制海马组织Bax mRNA的表达和促进Bcl-2mRNA表达,从而发挥其抗老年性痴呆神经细胞凋亡的作用。Objective: To observe the effect of TLJN Oral Solution on Bax and Bcl - 2 mRNA expression in brain tissue of amyloid- β1-40 injection into brain induced experimental AD rats in vivo. Methods: 140 SD rats were enrolled in the study, and they were equally randomly divided into blank controls group, sham controls group, negfive control group, positive control group and experiment group( 12mg · kg^-1 · d^-1, 24mg · kg^-1 · d^-1 , 48mg · kg^-1 · d^-1 ) ,A rat model of AD induced by amyloid- β1 -40 sterotaxis injection the bilateral Hippocampus. all rats were sacrificed at 5 week after treatment for 4 weeks. Bax and Bcl - 2 mRNA expression in brain tissue was assayed by real - time quantitative reverse transcription - PCR. Results : the Bax mRNA expression was up - regulate expression In model groups compared with the sham group and Bcl - 2 was down - regulate in that. In experiment groups the Bax mRNA expression being down - regulated and Bcl -2 being up -regulated compared with the model group ( P 〈0.05). Conclusion: amyloid -β1-40 injection the bilateral Hippocampus can up - regulate expression of Bax mRNA and down - regulate expression of Bcl - 2 in brain tissue. TLJN Oral Solution may down - regulate expression of Bax mRNA and up - regulate expression of Bcl - 2 mRNA.
