PTEN与MMP-7基因mRNA在胃癌中的表达及其临床意义

摘　　要：目的:探讨PTEN和MMP-7基因mRNA在胃癌中的表达规律及其临床意义。方法:应用逆转录-聚合酶链反应(RT-PCR)方法,对15例癌旁组织和46例胃癌组织中PTEN和MMP-7基因mRNA的表达进行分析。结果:胃癌组织中PTEN基因mRNA的表达明显降低,而MMP-7基因mRNA的表达却明显增加,分别为43.5%(20/46)和71.7%(33/46),P<0.01;且其表达与肿瘤的大小、分化程度及Lauren分型无关(P>0.05);而与胃癌的浸润深度(P<0.05)、是否有淋巴结转移(P<0.01)和临床分期(P<0.01)关系密切。PTEN和MMP-7mR-NA表达之间显著负相关(rs=-0.276,P<0.05);结论:PTEN、MMP-7基因参与了胃癌的发展过程,并与胃癌的部分生物学行为有关,可能成为胃癌诊断和判断其生物学特性的重要的分子生物学检测指标。Objective To study the expression of PTEN and MMP - 7 mRNA in human Gastric cancer and their clinical significance. Methods Reverse transcription polymerase chain reaction （ RT - PCR） was used to detect the expression of PTEN and MMP - 7 mRNA in 46 cases with gastric cancer. Results The expression of PTEN mRNA was significantly decreased in gastric cancer,while MMP -7 mRNA expression was increased （43.5% and 71.7% respectively,P 〈 0.01 ）. The expression of them was not related with the size,differentiation level and Lauren type of gastric cancer（ P 〉 0.05） , but related closely with the infiltration depth （ P 〈 0.05）, lymph node metastasis （ P 〈 0.01 ） and clinical stage （ P 〈 0. 01 ） The con＇elation of PTEN mRNA expression with MMP - 7 mRNA expression was significantiy negative （ r = - 0.276, P 〈 0. 05）. Conclusion PTEN and M MP- 7 expression may play important roles in the development of gastric cancer, and were related with partial biological behaviors of gastric cancer. The expression of PTEN and MMP - 7 mRNA may serve as important molecular biological indicators in diagnosing and predicting the biological behavior of gastric cancer.
