摘 要:目的研究反义c-myc寡核苷酸(c-mycASODN)对人骨肉瘤MG-63细胞增殖和凋亡的影响。方法设计c-mycASODN片段,转染入人骨肉瘤MG-63细胞,通过MTT检测、HE染色光镜观察、透射电镜超微结构及流式细胞仪(FCM)分析,观察和分析其对瘤细胞体外增殖、凋亡、细胞周期及c-myc基因蛋白的影响。结果MTT示c-mycASODN可抑制人骨肉瘤MG-63细胞的体外增殖,10.0μmol/L、作用48h效果最明显;形态学观察到典型瘤细胞凋亡特征;流式细胞仪分析证实反义c-myc寡核苷酸主要阻止瘤细胞进入S期,出现明显的凋亡峰,凋亡率达36.82%,并可抑制c-myc基因蛋白的表达。结论反义c-myc寡核苷酸可诱导人骨肉瘤MG-63细胞的凋亡,对瘤细胞的增殖有抑制作用。Objective To study the effects of c-myc antisense oligonucleotide on the proliferation and apoptosis of osteosarcoma cells (MG-63). Methods Designed c-myc antisense oligonucleotide fragment was transfected into human osteosarcoma MG-63 cell and its effects on the proliferation, apoptosis were observed by MTT method and HE stain and transmission electron microscope. Results It showed that c-myc antisense oligonucleotide was able to inhibit the proliferation of human osteosarcoma MG-63 cell at the doees of 10.0μmol/L and effected for forty-eight hours; Cells exhibited characteristic morphologic changes of apoptosis. FCM method checked human osteosarcoma MG-63 cell into the S phase induce apoptosis (the apoptosis rate could be to 36.82%) and inhibit the expression of c-myc gene protein. Conclusion c-myc antisense oligonucleotide is able to induce apoptosis of human osteosarcoma MG-63 cell and inhibit the proliferation of human osteosarcoma MG-63 cell.
