摘 要:目的通过检测胰腺癌组织HIF—1α、Bcl—XL的表达,探讨HIF—1α对胰腺癌细胞凋亡的影响。方法应用免疫组化方法检测41例胰腺癌和20例正常胰腺组织中HIF-1α、Bcl-XL的表达,TUNEL法检测细胞凋亡,RT—PCR和Western blot检测21例胰腺癌组织和10例正常胰腺中HIF-1α、Bcl-XLmRNA和蛋白的表达。结果正常胰腺组织均无HIF—1α、Bcl—XL蛋白和mRNA的表达。胰腺癌组织HIF—1α、Bcl—XL蛋白表达率分别为63.4%和87.8%,HIF—1αmRNA和Bcl—XL mRNA的表达率分别为71.4%和80.9%,两者的表达呈正相关(r=0.335,P=0.032),HIF-1α阳性表达的胰腺癌的凋亡指数较阴性表达者低[(1.32±0.59)%vs(1.79±0.77)%,P=0.034]。结论HIF-1α和Bcl—XL在胰腺癌组织中存在高表达。HIF—1α可能通过上调Bcl—XL的表达抑制胰腺癌细胞凋亡.在胰腺癌发生发展中起重要作用。Objective To investigate the expression of HIF-1α in pancreatic cancer tissue, and evaluate its effect on apoptosis of pancreatic cancer cells and explore the mechanism. Methods Expression of HIF-1α and Bcl-XL was detected by Immunohistochemistry in 41 cases of pancreatic cancer tissue and 20 cases of normal pancreas tissue. Expression of HIF-1α and Bcl-XL was detected by RT-PCR and Wistarn blot in 21 pancreatic cancer samples and 10 normal pancreases. Corre1αtion between the expression of HIF-1α and Bcl-XL and their re1αtionship with apoptotic index were statistically analyzed. Results These no expression of both HIF-1α and Bcl-XL in normal pancreatic tissue. The expression rate of HIF-1α and Bcl-XL was 63.4% and 87.8%, respectively, in pancreatic cancer tissues. Corre1αtion between HIF-1α and Bcl-XL expression levels was tested with Spearman rank corre1αtion, and the result was positive (r = 0. 335, P = 0. 032). Cancer tissues with positive HIF- 1α or Bcl-XL expression showed a lower score of apoptotic index than those with negative expression (P = 0. 034, P = 0. 008 respectively). The expression rate of HIF-1α mRNA and bcl-XL mRNA was 71.4% and 80.9%, respectively, in pancreatic cancer. Corre1αtion between them was positive also. Conclusions Expression of HIF-1α and Bcl-XL is strong in pancreatic cancer tissue. HIF-1α inhibits apoptosis, probably through up-regu1αting the expression of Bcl-XL.
