摘  要:目的:研究姜黄素对人类口腔鳞状细胞癌(oral squamous cell carcinom a,0SCC)生长增殖及相关蛋白表达的影响。方法:MTT法评定姜黄素细胞的毒性作用;形态学观察及流式细胞仪测定细胞凋亡;免疫组化法获得NF-KappaB和细胞周期蛋白D1(Cyclin D1)的表达变化。结果:5、10、20mg/L浓度的姜黄素具细胞毒性作用,流式反应显示≥5mg/L时24h即具有凋亡作用,NF-KappaB和Cyclin D1的表达随药物浓度增加而渐减弱。结论:姜黄素抑制鳞癌细胞生长,减少NF-KappaB和CyclinD1的表达,可作为口腔肿瘤的化学预防剂加以应用。Objective: To investigate the effects of curcumin on proliferation and related protein' s expression of human OSCC. Methods: The cytotoxic effect of curcumin was evaluated by MTT assay, apoptosis by a morphological assay, and flow cytometry and the expression of NF - Kappa B and Cyclin D1 by immunohistochemistry. Results: Cureumin concentrations of 5,10,20mg/L were cytotoxic for OSCC cells. Flow cytometry showed that concentrations ≥5mg/L could induce apotosis after 24h treatment. NF -Kappa B and Cyclin D1 decreased after the increasement of drug's density. Conclusion: The use of curcumin as an oral cavity cancer chemopreventive agent could have significant clinical impact via its ability to inhibit the growth of SCC cell. This might be associated with the decreased NF - KappaB and CyclinD1 expression.
