摘  要:目的研究C3d分子对多表位DNA疫苗体液免疫应答的增强作用。方法通过PCR方法合成多表位HME基因,分别插入真核表达载体pcDNA3.1和pcDNA3.1-3C3d,构建多表位DNA疫苗pcDNA3.1-HME和pcDNA3.1-HME-3C3d,经肌肉注射免疫小鼠,ELISA检测小鼠特异性抗体水平。结果经免疫的小鼠均能产生针对各表位的特异性IgG抗体,且pcDNA3.1-HME-3C3d免疫组小鼠的抗体水平明显高于pcDNA3.1-HME免疫组。结论C3d可增强多表位DNA疫苗的体液免疫应答水平。Objective To explore the enhancement of C3d molecule to the humoral immune response induced by polytope DNA vaccine. Methods Synthesize polytopo HME gene by PCR and insert into expression vectors pcDNA3.1 and pcDNA3.1-3C3d respectively. Inject i. m. mice with the constructed recombinant plasmids pcDNA3.1-HME and pcDNA3.1-HME-3C3d respectively and determine the serum IgG by ELISA. Results The IgG antibodies specific to various epitopes were induced in all the immunized mice. However,the IgG level of mice immunized with pcDNA3.1-HME-3C3d was significantly higher than that with pcDNA3. 1-HME. Conclusion C3d enhanced the humoral immune response to polytope DNA vaccine.
