摘　　要：目的探讨人巨细胞病毒(HCMV)感染和血管紧张素受体Ⅱ型(AngⅡ)对人脑动脉血管平滑肌细胞(vascular smooth muscle cells,VSMCs)中SM22α表达的影响。方法采用RT-PCR等技术观察VSMCs在更昔洛韦(GCV)、AngⅡ以及AT1和AT2受体拮抗剂(valsartan与P123319)作用下其SM22α的表达。结果病毒感染5 d VSMCs中SM22α基因表达量较感染3 d和1 d时明显上调。经AngⅡ诱导1 h和6 h后,VSMCs中SM22α相对表达量较未诱导组明显上调。AngⅡ+valsartan组SM22α相对表达量高于AngⅡ诱导组,AngⅡ+P123319组SM22α相对表达量低于AngⅡ诱导组;AngⅡ诱导12 h和24 h后,3组间SM22α相对表达量差异无统计学意义。结论HCMV感染可使VSMCs中SM22α基因表达上调,AngⅡ对VSMCs的早期作用可能通过AT2受体途径激活SM22α基因表达。Objective To study the pathogenesis of atherosclerosis by the expression of SM22α in smooth muscle cells of human brain artery after HCMV infection and Ang Ⅱ treatment. Methods Using of HCMV AD169 strain infected VSMCs in vitro and to establish the cell model of viral infection. The expression of SM22α in VSMCs was undergone HCMV and was treated with Ang Ⅱ. RT-PCR techniques were utilized for this research project. Results SM22α expression was showed up regulation in the HCMV infection group at 5 days after infection, and it was much higher than that at 3 days and 1 day. Up regulation of SM22α expression was also found in Ang Ⅱ group that was treated with AT1 receptor antagonist （valsartan） at 1 h and 6 h after Ang Ⅱ treatment, but down regulation of SM22α in the treated group with AT2 receptor antagonist （P123319）. It was no significant discrepancy between after Ang Ⅱtreatment at 12 h or 24 h in all groups. Conclusions HCMV infection group showed up regulation of SM22α expression in VSMCs in vitro. 5The up regulation o5 SM22α mRNA level was detected after Ang Ⅱ treatment, it might be activated via a pathway of AT2 receptor treated by Ang Ⅱ at the early time point.
