摘 要:目的:评价吡喃糖氧化酶(PROD)对不同的糖底物、甘油和乙二醇的催化作用。方法:应用相同酶试剂和固定参数对12种糖、甘油和乙二醇进行催化率实验。结果:将其对葡萄糖的催化作用定为100%时,PROD对单糖中的山梨糖催化率最高(87.51%),其次是木糖(83.17%)和1,5-脱水-D-山梨醇(82.32%),半乳糖为40.69%,对果糖、甘梨醇、山梨醇几乎无催化作用;PROD对双糖中的麦芽糖催化率为3.19%,对乳糖、蔗糖、纤维二糖几乎无催化作用;对同等浓度的非糖类物质甘油和乙二醇的催化作用很小,但是对高浓度的甘油和乙二醇(应用浓度在2.2%~3.3%)催化活性很高。结论:建立1,5-脱水-D-山梨醇测定方法前应进行PROD对不同糖底物、甘油和乙二醇的特异性催化实验。Objective: To evaluate the catalyzing effects of pyranose oxidase (PROD) on different sugar substrates, glycerol and ethylene glycol. Methods: The same working enzymatic reagents were used for evaluating the catalyzing specificity to all 12 sugar substrates, glycerol and ethylene glycol with the same test parameters. Results: The effect of catalyzing for glucose was de- termined as 100%. The PROD reagent had marked catalyzing effects for L-sorbose (87.51%), D-xylose (83.17%), 1,5-anhydro- D-glucitol (82.32%) and D-galactose (40.69%). There was little effect for D-fructose, mannitol and sorbitol in monosaccharides. The PROD had catalyzing effects for maltose(3.19%) but little effect for a-lactose, sucrose and cellobiose. The PROD had a little effect for glycerol and ethylene glycol with the same concentration but higher catalyzing effects for high concentration of glycerol and ethylene glycol (2.2%-3.3%). Conclusion: Before a new assay is set up the catalyzing specificity of the enzyme for its substrate should be evaluated.
