摘 要:研究了全长9.8~10.4cm,体重11.0~11.7g的鮸鱼幼鱼鳃丝Na+-K+-ATPase的性质。结果表明,鮸鱼鳃丝Na+-K+-ATPase活力的最适宜温度为30~35℃;pH<6.0时酶活力被抑制,pH>8.0则活力急剧下降,其最适宜pH为7~8;酶活力随着反应时间的增加而逐渐升高,且上升的趋势变慢;底物浓度小于9mmol/L时酶活力随着浓度的增加而上升,在高浓度下出现酶活力被抑制的现象;应介质中Na+、K+和Mg2+浓度的增加均出现峰值的变化,Na+、K+和Mg2+的最适宜的浓度分别为140~170,25~35和5~7mmol/L,K+和Mg2+在低浓度下酶活力抑制现象显著;乌本苷浓度为2.5mmol/L时酶活力完全被抑制。The characterization of gill Na^+-K^+-ATPase in Miichthys miiuy which span were 9.8-10.4 cm and avoirdupois were 11.0-11.7 g was studied. The results show that the optimal reaction temperature is 30- 35℃. The optimal pH is 7.0-8.0, while pH is less than 6.0 or higher than 8.0 the enzyme activity drops rapidly. In the range of reaction time, the gill Na^+-K^+-ATPase activity has arising trend along with the time prolonging but leveling off gradually. When the ATP-Na2 concentration of this experiment is less than 9.0 mmol/L the gill Na^+-K^+-ATPase activity has arising trend along with the concentration accessorial, but when the concentration is higher than 9.0 mmol/L the Na^+-K^+-ATPase activity will has been inhibited. The optimal concen- trations of Na^+, K^+ and Mg^2+ in the medium arel40-170 mmol/L, 25-35 mmol/L and 5-7 mmol/L respectively when the concentrations of K^+ or Mg^2+ are low, the gill Na^+-K^+-ATPase shows very low activity. At 2.5 mmol/L concentration of ouabain, the enzyme activity will been entirely inhibited.
